清香散对肝癌前病变大鼠模型病理形态及转化生长因子-β1表达的影响

Effect of Qingxiang powder on pathological morphology and TGF-β expression in rat models of liver precancerous lesion

摘要 目的分析清香散对肝癌前病变大鼠模型病理形态及转化生长因子-β1(TGF-β1)表达的影响。方法选取80只健康雄性无特定病原体级Wistar大鼠随机分为对照组、模型组、清香散大剂量组和小剂量组,每组各20只。使用二乙基亚硝胺(DEN)诱导肝癌癌前病变模型后,小剂量组使用1 kg/L的清香散灌胃,大剂量组为3 kg/L。实验28周后采用免疫组化法和HE染色法对比各组大鼠肝组织的病理形态变化、肝功能及TGF-β1表达。结果对照组无大鼠死亡,模型组、大剂量组和小剂量组大鼠的存活率分别为55.0%(11/20)、95.0%(19/20)和85.0%(17/20),大剂量组和小剂量组的存活率均高于模型组(χ^(2)=8.533,P=0.003;χ^(2)=4.286,P=0.038),但大剂量组与小剂量组大鼠的存活率差异无统计学意义,χ^(2)=1.111,P=0.292。单因素ANOVA分析结果显示,各组大鼠的肝功能指标比较差异有统计学意义,F值分别为118.560、102.027和164.342,均P<0.001。采用LSD法进行多重比较后显示,模型组、大剂量组和小剂量组大鼠的各项肝功能表达水平均高于对照组(P<0.05),大剂量组和小剂量组均低于模型组(P<0.05),大剂量组低于小剂量组,P<0.05。对照组TGF-β1表达为54.51±4.01,模型组为37.08±3.25,大剂量组为50.17±3.07,小剂量组为45.59±3.38,差异有统计学意义,F=93.726,P<0.001。结论清香散可以通过提高肝组织的TGF-β1表达,改善DEN所诱导肝癌癌前病变大鼠肝脏组织的病理形态,提高大鼠的肝功能。 Objective To analyze the effects of Qingxiang powder on the pathological morphology and the expression of transforming growth factor-β1(TGF-β1)in a rat model of liver precancerous lesions.Methods Eighty healthy male SPF Wistar rats were selected as experimental animals,and they were randomly divided into control group,model group,Qingxiang powder high-dose group and low-dose group,each with 20rats.Diethylnitrosamine(DEN)was used to induce the liver cancer precancerous lesion model.The low-dose group was given 1kg/L Qingxiang powder by gavage,and the high-dose group was given 3kg/L Qingxiang powder by gavage.After 28weeks of the experiment,immunohistochemistry and HE staining were used to compare the pathological changes,liver function and TGF-β1expression of the liver tissues of rats in each group.Results No rats died in the control group.The survival rates of the model group,high-dose group and low-dose group were 55.0%(11/20),95.0%(19/20)and 85.0%(17/20),respectively.The survival rate of the dose group and the low-dose group was higher than that of the model group(χ^(2)=8.533,P=0.003;χ^(2)=4.286,P=0.038),but there was no difference in statistical significance in the survival rate of rats between the high-dose group and the lowdose group(χ^(2)=1.111,P=0.292).The results of one-way ANOVA analysis showed that the liver function indexes of rats in each group were significantly different(Fvalues were 118.560,102.027and 164.342,all P<0.001),and the LSD method multiple comparisons showed that the expression levels of various liver functions of the model group,high-dose group and low-dose group were higher than those of the control group(P<0.05),the expression levels in the high-dose group and low-dose group were lower than those in the model group(P<0.05),the expression levels in the high-dose group was lower than those in the low-dose group(P<0.05).The expressions of TGF-β1in the control group,model group,high-dose group and low-dose group were 54.51±4.01,37.08±3.25,50.17±3.07and 45.59±3.38,respectively(F=93.726,P<0.001).Conclusion Qingxiang powder can improve the pathological morphology of liver tissue in rats with precancerous lesions of liver cancer induced by DEN by increasing the expression of TGF-β1in liver tissue,and improve the liver function of rats.
