菟丝子对幼龄大鼠慢性腹泻中肠道炎症、肠道菌群及肠黏膜微观结构变化的影响 被引量:10

摘要 目的探讨菟丝子对幼龄大鼠慢性腹泻中肠道炎症、肠道菌群及肠黏膜微观结构变化的影响。方法将SD大鼠分为假手术组(sham组)、模型组(model组)、菟丝子低剂量组(SC-L组)、菟丝子高剂量组(SC-H组)。记录各组大鼠体重变化、排便量和湿粪率。采用试剂盒检测各组大鼠血清D-木糖含量;ELISA检测各组大鼠血清肿瘤坏死因子-α(TNF-α)、白细胞介素-1β(IL-1β)和白细胞介素-10(IL-10)的含量;HE染色观察各组大鼠结肠组织病理变化;电镜观察各组大鼠肠黏膜微观结构变化;qRT-PCR检测各组大鼠粪便中双歧杆菌、乳酸杆菌和大肠杆菌的含量。结果与sham组比较,model组大鼠体重减少,排便量和湿粪率增加;血清D-木糖、IL-10含量降低,TNF-α、IL-1β含量增加;大鼠结肠组织受损;粪便中双歧杆菌、乳酸杆菌相对数量减少,大肠杆菌相对数量增加。与model组相比,SC-L组、SC-H组大鼠体重增加,排便量和湿粪率降低;血清D-木糖、IL-10含量升高,TNF-α、IL-1β含量降低;受损的大鼠结肠组织上皮和黏膜得到修复;粪便中(SC-H组)双歧杆菌、乳酸杆菌相对数量增加、大肠杆菌相对数量降低。差异均有统计学意义(P<0.05)。结论菟丝子减轻大鼠的腹泻症状可能与减轻炎性因子的表达、改善肠道菌群数量及肠黏膜损伤有关。 Objective To explore the effect of Semen cuscutae on intestinal inflammation,intestinal flora and microstructure of intestinal mucosa in young rats with chronic diarrhea.Methods The SD rats were divided into 4 groups:sham operation group(sham group),model group(model group),Semen cuscutae low-dose group(SC-L group)and Semen cuscutae high-dose group(SC-H group).The weight change,defecation volume and wet fecal rate of rats in each group were recorded.The kit was used to detect the serum D-xylose content of each group of rats;ELISA was used to detect the levels of serum tumor necrosis factor-α(TNF-α),interleukin-1β(IL-1β)and interleukin-10(IL-10)in each group of rats;HE staining was used to observe the pathological changes of the colon tissue of rats in each group;electronic microscope was used to observe the microstructure of intestinal mucosa of rats in each group;qRT-PCR was used to detect the contents of Bifidobacterium,Lactobacillus and Escherichia coli in the feces of each group.Results Compared with sham group,the body weight of rats in model group was decreased,while the defecation volume and rate of wet feces was increased(P<0.01);the contents of serum D-xylose and IL-10 were decreased(P<0.01),while the contents of serum TNF-αand IL-1βwere increased(P<0.01);the colon tissue was damaged;the relative number of Bifidobacterium and Lactobacillus in rats′feces was increased(P<0.01),while that of E.coli was decreased(P<0.01).Compared with model group,the body weight of rats in SC-L group and SC-H group was increased,while the defecation volume and rate of wet feces was decreased(P<0.05);the contents of serum D-xylose and IL-10 were increased(P<0.05),while the contents of serum TNF-αand IL-1βwere decreased(P<0.05);The epithelium and mucous membrane of the damaged rat colon tissue was repaired;the relative number of Bifidobacterium and Lactobacillus in rats′feces was decreased(P<0.05),while that of E.coli was increased(P<0.05).Conclusion Semen cuscutae alleviates diarrhea symptoms in rats,which may be related to reducing the expression of inflammatory factors,improving the number of intestinal flora and intestinal mucosal damage.
