猪源副流感病毒5型SH毒株全基因组测序及分析

摘要 目的对猪源副流感病毒5型(parainfluenza virus 5,PIV5)SH毒株进行全基因组测序及分析。方法根据GenBank中登录的猪源PIV5基因组序列设计10对引物,采用RT-PCR法对各片段进行扩增、测序,并对获得的PIV5-SH株全基因组序列进行遗传进化性分析。结果 PIV5-SH毒株基因组长15 246 nt,与传统毒株PIV5-W3A核苷酸序列同源性为98.7%。基于全基因组构建的遗传进化树显示,PIV5-SH毒株与韩国犬源PIV5(1168-1)、中国孟加拉虎源PIV5(PIV5-HMZ)、中国东北虎源PIV5(PIV5-SR)及中国小熊猫源PIV5(ZJQ-221)具有较近的亲缘关系,与传统毒株W3A亲缘关系较远。进一步以PIV5 NP、F及HN基因构建遗传进化树显示,PIV5-SH毒株与PIV5 1168-1、PIV5-HMZ、PIV5-SR、PIV5 ZIQ-221和PIV5-CAN毒株亲缘关系近。结论 PIV5不同分离株基因差异较小,表现出一定的遗传稳定性。 Objective To sequence and analyze the complete genome of porcine parainfluenza virus 5 strain SH(PIV5-SH).Methods Ten pairs of primers were designed according to the genome sequence of PIV5 in GenBank for amplification of various fragments by RT-PCR.The obtained complete genome of PIV5-SH was sequenced and subjected to phylogenetic analysis.Results The complete genome of PIV5-SH was at a length of 15 246 nt,of which the homology of nucleotide sequence was 98.7% to that of PIV5-W3 A,a prototype PIV5 strain.Phylogenetic tree based on complete genome showed close relationship of PIV5-SH to PIV5 strain 1168-1 from canine in the Republic of Korea,PIV5 strain HMZ from Panthera tigris tigris in China,PIV5 strain SR from Panthera tigris altaica in China and PIV5 strain ZJQ-221 from Ailurus fulgens in China,while showed far relationship to PIV5-W3 A.However,the phylogenetic tree based on NP,F and HN genes showed close relationship of PIV5-SH to PIV5 1168-1,PIV5-HMZ,PIV5-SR,PIV5 ZJQ-221 and PIV5-CAN.Conclusion Various PIV5 isolates showed little difference in genes,indicating a certain genetic stability.
