CXC亚家族趋化因子13在乳腺癌组织中表达及其调控网络的生物信息学分析 被引量:1

摘要 目的通过生物信息学方法分析CXC亚家族趋化因子13(CXC subfamily chemokine 13,CXCL13)在乳腺癌组织中的表达及其调控网络。方法利用多种在线数据库分析CXCL13在乳腺癌组织中的表达、基因变化及相关的功能网络,筛选CXCL13相关的差异表达基因,进行基因本体论及京都基因与基因组百科全书信号通路分析,同时分析CXCL13正相关基因集在乳腺癌中的激酶、转录因子及miRNA类型的靶点网络。结果 CXCL13在乳腺癌组织中过表达(P<0.01);扩增是CXCL13在乳腺癌中最常见的突变类型;CXCL13基因的变化对G蛋白偶联受体的激活、信号传递及T细胞激活等多个过程具有深远的影响;CXCL13与几种肿瘤相关激酶(如FYN)、转录因子(如NF-κB)及miRNA(如miRNA-146a)特异相关。结论 CXCL13可能是乳腺癌治疗中一个潜在的靶点,为进一步研究CXCL13在肿瘤发生中的作用奠定了基础。 Objective To analyze the expression and regulatory network of CXC subfamily chemokine 13(CXCL13) in breast cancer tissue by bioinformatic method.Methods The expression of CXCL13 in breast cancer was analyzed by using Oncomine database,while the genetic change and related functional networks by using cBioPortal database.The differentially expressed genes were screened by using LinkedOmics database and analyzed for gene ontology,kyotoencyclopedia of genes and genome pathway.Meanwhile,differential gene enrichment analysis of target networks for detection of kinasee,transcription factors and miRNAs was performed.Results CXCL13 was over-expressed in breast cancer tissue(P<0.01),while the most common mutation of CXCL13 in breast cancer cells was amplification mutation.The changes in CXCL13 gene showed significant effect on multiple processes such as activation of G-protein coupled receptor,signal transduction and T cell activation.CXCL13 was specifically associated with several tumor-associated kinases(such as FYN),transcription factors(such as NF-κB)and miRNAs(such as miRNA-146 a).Conclusion CXCL13 might be a potential target in therapy of breast cancer,which laid a foundation of further study on the role of CXCL13 in tumorigenesis.
