丁苯酞对大鼠冠状动脉微栓塞后心肌炎症反应的影响 被引量:1

Effects of Butylphthalide on Myocardial Inflammatory Response after Coronary Microembolization in Rats

摘要 目的观察丁苯酞(NBP)对大鼠冠状动脉微栓塞(CME)后心肌炎症反应的影响。方法将36只无特定病原体(SPF)级雄性SD大鼠随机分为CME组、假手术组(Sham组)、CME+丁苯酞预处理组(CME+NBP组),每组12只。CME+NBP组术前给予NBP 80 mg/kg灌胃7 d。术后6 h检测各组大鼠心功能,苏木精-伊红(HE)染色观察心肌组织病理改变,逆转录-聚合酶链式反应(RT-PCR)检测心肌组织白细胞介素1β(IL-1β)mRNA、肿瘤坏死因子α(TNF-α)mRNA相对表达水平,免疫印迹法(Western Blotting)检测心肌组织IL-1β、TNF-α蛋白相对表达水平。结果与Sham组比较,CME组大鼠左室射血分数(LVEF)和短轴缩短率(FS)降低(P<0.05),左室舒张末期内径(LVEDD)和左室收缩末期内径(LVESD)增加(P<0.05);与CME组比较,CME+NBP组大鼠LVEF、FS升高(P<0.05),而LVEDD、LVESD降低(P<0.05)。HE染色显示:CME+NBP组心肌细胞坏死及白细胞浸润较CME组减轻。与Sham组比较,CME组IL-1β、TNF-αmRNA及蛋白相对表达水平升高(P<0.05);与CME组比较,CME+NBP组IL-1β、TNF-αmRNA及蛋白相对表达水平降低(P<0.05)。结论NBP预处理可减轻大鼠CME后心肌炎症反应并保护心功能。 Objective To observe the effect of butylphthalide(NBP)on myocardial inflammatory response after coronary microembolization(CME)in rats.Methods Thirty-six SPF male SD rats were randomly divided into control group(Sham group),coronary microembolization group(CME group),CME+NBP group,with 12 rats in each group.CME+NBP group was given 80 mg/kg NBP for 7 days before operation.After 6 h,cardiac function was detected.Pathological changes of myocardial tissue were observed by hematoxylin-eosin(HE)staining.The relative expression levels of IL-1βmRNA and TNF-αmRNA in myocardial tissue were detected by reverse transcription-polymerase chain reaction(RT-PCR).The expression levels of IL-1βand TNF-αin myocardial tissue were detected by Western Blotting.Results Compared with the Sham group,left ventricular ejection fraction(LVEF)and short axis shortening rate(FS)in the CME group were decreased(P<0.05),and left ventricular end diastolic diameter(LVEDD)and left ventricular end systolic diameter(LVESD)were increased(P<0.05).Compared with CME group,LVEF and FS in the CME+NBP group were increased(P<0.05),while LVEDD and LVESD were decreased(P<0.05).HE staining showed that myocardial cell necrosis and leukocyte infiltration were less in the CME+NBP group than those in the CME group.Compared with the Sham group,the mRNA and protein relative expression levels of IL-1βand TNF-αmRNA and protein in the CME group were increased(P<0.05).Compared with the CME group,the relative expression levels of IL-1βand TNF-αmRNA and protein in the CME+NBP group were decreased(P<0.05).Conclusion NBP pretreatment could reduce myocardial inflammatory response and protect cardiac function in rats after CME,which might improve myocardial injury after CME by reducing myocardial inflammatory response.
