摘要
通过合成一种水溶性双核铜(Ⅱ)配合物[Cu_(2)(BPH)_(2)Cl_(4)]·CH_(3)OH(BPH为1-苯甲酰-1-苯肼),利用元素分析、红外光谱和X-射线单晶衍射等方法对其进行表征.从配合物的单晶结构可以看出,两个[Cu(BPH)Cl]单元通过两个氯离子桥接,每个Cu(Ⅱ)离子都为五配位的变形四方锥构型.利用多种光谱分析方法研究配合物与DNA的相互作用,结果表明该配合物可能是通过沟槽结合方式与DNA结合.用凝胶电泳方法研究该配合物对超螺旋pBR322DNA的切割作用,在H_(2)O_(2)存在的情况下,该配合物能有效切割DNA,其作用机理为氧化切割,单线态氧与金属离子可能对切割起关键作用.
A water soluble binuclear Cu(Ⅱ) complex[Cu2(BPH)2Cl4]·CH3OH (BPH is 1-benzoyl-1-phenylhydrazine) was synthesized and characterized by elemental analysis,infrared spectroscopy and single crystal X-ray crystallography.In the complex,two[Cu(BPH)Cl]units are bridged by two chloride ions,and each Cu(Ⅱ)ion displays a five-coordinate slightly distorted square-pyramid structure.Multi-spectroscopic DNA interactions studies indicated that the complex may bind to DNA via groove binding modes.The cleavage effect of the complex on supercoiled pBR322 DNA was studied by gel electrophoresis method.The results show that the complex could cleave DNA effectively in the presence of H2O2via an oxidative mechanism.The singlet oxygen and metal ions may play crucial roles in the DNA cleavage process.
