双佐剂新型冠状病毒灭活疫苗免疫原性及中和效价的测定 被引量:2

Determination of Immunogenicity and Neutralization Titer of the Inactivated SARS-CoV-2 Vaccine with Dual Adjuvant

摘要 在Balb/c小鼠模型上,评价双佐剂新型冠状病毒灭活疫苗的免疫效果及中和抗体水平.新型冠状病毒经Vero细胞培养、收获、浓缩、灭活、纯化等一系列步骤,制成灭活疫苗原液,以不同的配方剂量添加CpG佐剂和氢氧化铝佐剂后,免疫Balb/C小鼠,然后检测小鼠血清特异性抗体水平及中和抗体效价水平.结果表明:低、中剂量新冠抗原添加低、中剂量的CpG佐剂后,可以有效地提高S蛋白特异性抗体效价水平及中和抗体效价水平,效果远大于单独铝佐剂组;但CpG剂量如果过高,会有抑制效果.添加CpG佐剂后,可以使新冠病毒灭活抗原在较低的剂量下,就能产生较高水平的S蛋白特异性抗体效价及中和抗体效价. This article valuates the immune effect and neutralization titer of the inactivated SARS-CoV-2 vaccine with dual adjuvant on the Balb/C mouse model.An inactivated vaccine bulk was prepared through a series of steps,which include culturing coronavirus in VERO cells,harvesting,concentrating,inactivating,and purifing.Then,CpG adjuvant and aluminum hydroxide adjuvant of different dosages were added and the Balb/C mouse was immunized.Finally,the level of specific antibody and neutralizing antibody in the serum of the mouse was detected.The results show that low and medium doses of inactivated SARS-CoV-2 antigens with addition of low and medium doses of CpG adjuvants can increase the titers of S protein specific antibodies and neutralizing antibodies effectively,andthe effect is much greater than that of the single aluminum adjuvant group;but if the CpG dose is too high,there will be an inhibitory effect.The inactivated SARS-CoV-2 antigen can produce a higher level of S protein specific antibody titers and neutralizing antibody titers at a lower dose after adding CpG adjuvant.
