倒心盾翅藤总皂苷提取工艺优化 被引量:2

Optimization of Extraction Technology for Total Saponins in Aspidopterys Obcordata Vines

摘要 甾体总皂苷为倒心盾翅藤抗肾结石的活性成分,该研究选用菝葜皂苷元为对照品,以1%茴香醛-硫酸作为显色剂,建立倒心盾翅藤甾体总皂苷含量测定方法。分别评价了乙醇体积分数、液料比、提取温度、提取时间对甾体总皂苷得率的影响,并且进一步通过响应曲面法优选提取工艺为乙醇体积分数50%、液料比15、提取温度100℃、提取时间3 h,甾体总皂苷得率为2.37%±0.19%。该提取工艺稳定可行,所得甾体总皂苷生物活性明确。 The extracting technology of Total Saponins from Aspidopterys obcordata vines was optimized.Sasaponin was selected as a reference,and 1%anisaldehyde-sulfuric acid was used as a coloring agent to establish a quantitative method for the determination of total saponins of Aspidopterys obcordata vines.The effects of ethanol volume fraction,liquid-to-material ratio,extraction temperature,and extraction time on the yield of total steroidal saponins were respectively evaluated,and the extraction process was further optimized by response surface method as ethanol volume fraction of 50%,liquid-to-material ratio of 15 and extraction temperature of 100℃,extraction time 3h,the yield of total steroidal saponin was 2.37%±0.19%.This extraction method can obtain total saponins with clear biological activity,accounting for 2.3%of the quality of medicinal materials,which is stable and feasible.
