TRAF6、Nectin-4在食管癌组织中的表达水平及临床意义

Expression Level and Clinical Significance of TRAF6 and Nectin-4 in Tissues of Patients with Esophageal Cancer

目的探讨肿瘤坏死因子受体相关因子6(TRAF6)、连接蛋白4(Nectin-4)在食管癌组织中的表达水平及临床意义。方法采用免疫组织化学法,应用RT-PCR技术检测91例食管癌患者肿瘤标本中TRAF6、Nectin-4及mRNA的表达水平,分析其与患者化疗效果的关系,采用Kaplan-Meier生存曲线分析其与患者预后的关系。结果食管癌组织中TRAF6、Nectin-4及mRNA表达水平明显高于癌旁组织(P<0.001);TRAF6、Nectin-4高表达与患者年龄、性别、肿瘤类型及发生部位无关(P>0.05),与患者有无淋巴结转移及肿瘤分期相关(P<0.05);化疗后各效果组组织中TRAF6、Nectin-4 mRNA水平具有统计学差异(P<0.001),CR组、PR组患者组织中TRAF6、Nectin-4 mRNA水平明显低于PD组患者(P<0.05);组织中TRAF6、Nectin-4高表达患者3年生存率明显低于低表达患者(P<0.05)。结论TRAF6、Nectin-4在食管癌组织中表达水平明显升高,且与患者化疗效果和预后相关,可能作为预测食管癌化疗效果和预后的相关指标发挥作用。

Objective To explore the expression level and clinical significance of Tumor necrosis factor receptor-associated factor 6(TRAF6)and Nectin-4in tissues of patients with esophageal cancer.Methods Immunohistochemistry and RT-PCR were used to detect the expression levels of TRAF6,Nectin-4 and mRNA in the tumor specimens of 91 patients with esophageal cancer,and analyze their relationship with chemotherapy Effect relationship,using Kaplan-Meier survival curve to analyze its relationship with patient prognosis.Results The expression levels of TRAF6,Nectin-4 and mRNA in esophageal cancer tissues were significantly higher than those in adjacent tissues(P<0.001);the high expression of TRAF6 and Nectin-4 was not related to the patient’s age,gender,tumor type and location(P>0.05).It was related to whether had lymph node metastasis and tumor stage(P<0.05);after chemotherapy,the levels of TRAF6 and Nectin-4 mRNA in the tissues of each effect group were statistically different(P<0.001),and TRAF6,Nectin-4 mRNA levels of the CR and PR groups were significantly lower than that of PD patients(P<0.05);the 3-year survival rate of patients with high expression of TRAF6 and Nectin-4 was significantly lower than that of patients with low expression of TRAF6 and Nectin-4(P<0.05).Conclusion The expression levels of TRAF6 and Nectin-4 in esophageal cancer tissues are significantly increased,and are related to the chemotherapy effect and prognosis of patients,and may play a role in predicting the chemotherapy effect and prognosis of esophageal cancer.
