INSIG1基因多态性与成年人肥胖的相关性研究

摘要 目的探讨INSIG1基因的单核苷酸多态性(SNP)与成年人肥胖的相关性。方法本研究为病例对照研究,包括516名肥胖受试者(病例组)和463名年龄和性别匹配的正常体重对照者(对照组)。使用TaqManSNP基因分型分析对rs2721进行基因分型。结果rs2721基因型分布,显性模型(GT/TT vs GG),隐性模型(TT vs GT/TT)在病例组和对照组之间的差异有统计学意义(分别为P=0.008,P=0.005和P=0.035)。校正混杂因素后,rs2721 GT基因型的显性模型(GT/TT vs GG)仍与肥胖明显相关(OR=1.393,95%CI=1.047~1.853,P=0.023)。rs2721 GT/TT基因型的TG水平明显高于GG基因型(P<0.05)。结论新疆成年人INSIG1基因rs2721位点与肥胖相关。rs2721的GT/TT基因型或T等位基因明显增加肥胖风险。 Objective Obesity is a major risk factor for chronic and metabolic diseases.Insulin inducible gene 1(IN⁃SIG1)plays an important role in cholesterol and triglyceride metabolism.In this study,we examined whether the sin⁃gle-nucleotide polymorphism INSIG1 gene was associated with obesity or not in Xinjiang adults.Methods A case-con⁃trol study was designed enrolling 516 obese patients and 463 control subjects matched with similar age and sex distri⁃bution.RS2721 was genotyped by TaqMan single nueleotide polymorphism(SNP)genotyping analysis.Results RS2721 genotype distribution,dominant model(GT+TT vs GG),recessive model(TT vs GT+GG)in obese patients and control group were significantly different(P=0.008,P=0.005 and P=0.035,respectively).After adjustment for con⁃founding factors,the dominant model of RS2721 GT genotype(GT+TT vs GG)was still significantly associated with obesity(OR=1.393,95%CI:1.047-1.853,P=0.023).The TG level of RS2721 GT/TT genotype was significantly higher than that of GG genotype(P<0.05).Conclusion The RS2721 locus of INSIG1 gene is associated with obesity in Xinji⁃ang adults.The GT/TT genotype or t allele of RS2721 significantly increased the risk of obesity.
