IDOL基因多态性与成年人肥胖的相关性研究

Study on the association between IDOL gene polymorphism and obesity in adult population

目的探讨人类诱导型低密度脂蛋白受体降解蛋白(IDOL)基因多态性与成年人肥胖的相关性。方法采用病例对照研究,选择2013年8月—2019年10月期间因心血管疾病在新疆医科大学第一附属医院心脏中心住院治疗的患者1172例,根据肥胖诊断标准分为肥胖组(n=572)和对照组(n=600),采用改进的多重连接检测反应(iMLDR)方法对诱导型低密度脂蛋白受体降解蛋白IDOL基因rs2072783、rs9370867和rs9370867位点进行基因型分型。结果诱导型低密度脂蛋白受体降解蛋白IDOL基因SNP1(rs2072783)在肥胖组及对照组中基因型分布频率、等位基因频率、显性模型(AA vs AG+GG)、隐形模型(GG vs AG+AA)的分布频率存在统计学差异。SNP2(rs9370867)在肥胖组及对照组中基因型分布频率、等位基因频率、显性模型(AA vs AG+GG)、相加模型(AG vs AA+GG)的分布频率存在统计学差异(P<0.05)。校正混杂因素后,rs2072783位点的隐形模型(GG vs AA+AG)和rs9370867位点的相加模型(AG vs AA+GG)为肥胖患者的危险因素[(rs2072783:OR=1.482,P=0.032,95%CI:1.034-2.121);(rs9370867:OR=1.849,P=0.032,95%CI:1.034-2.121)]。结论诱导型低密度脂蛋白受体降解蛋白IDOL基因多态性与成年人肥胖患病具有相关性。IDOL基因rs9370867和rs2072783位点G等位基因的人群比携带A等位基因的人罹患肥胖的可能性更高,诱导型低密度脂蛋白受体降解蛋白IDOL基因rs9370867位点AG基因型及rs2072783位点的GG基因型是肥胖的易感因素。

Objective To investigate the relationship between IDOL gene polymorphism and obesity in adult population.Methods A case-control study was designed and patients with cardiovascular diseases who were hospitalized at the Heart Center of Xinjiang Medical University Affiliated First Hospital from August 2013 to October 2019 were enrolled into this study.Case and control groups were divided according to obesity diagnostic criteria and genotyped for IDOL gene rs2072783,rs9370867 and rs9370867 loci using the modified multiplex linkage detection response(iMLDR)method.Results For the IDOL gene rs2072783 variant,the distribution of genotypes,dominant model(AA vs.AG+GG)and recessive model(GG vs.AG+AA)were significantly different between the case group and the control group(P<0.001).For the IDOL gene rs9370867 variant,the distribution of genotypes,dominant model(AA vs.AG+GG)and additive model(AG vs.AA+GG)were significantly different between the case group and the control group(P<0.001).Logistic regression analysis showed that the rs2072783 and rs9370867 polymorphism of IDOL gene were an in⁃dependent risk factor for obesity[(rs2072783:OR=1.482,P=0.032,95%CI:1.034-2.121);(rs9370867:OR=1.849,P=0.032,95%CI:1.034-2.121)].Conclusion IDOL gene rs2072783 and rs9370867 variants may be associated with obe⁃sity in Xinjiang adult population.
