摘要
目的探讨microRNA-24(miR-24)在阻塞性睡眠呼吸暂停低通气综合征(OSAHS)患者血清中的表达变化及其与胰岛β细胞功能受损状况的研究。方法研究纳入正常身体质量指数(BMI)的健康志愿者80例,正常BMI的OSAHS患者80例,超重或肥胖的OSAHS患者80例。采用多导睡眠监测仪监测获得受试者夜间睡眠状况。采用实时定量PCR实验检测患者血清miR-24水平;酶联免疫吸附实验检测血清C肽水平;全自动生化分析仪测定血糖水平。结果与正常组相比,OSAHS患者血清miR-24水平升高(P<0.001),而超重或肥胖可导致miR-24水平进一步升高(P<0.001)。OSAHS患者胰岛β细胞功能紊乱,表现为服糖后30,60,120 min后C肽水平及空腹和餐后血糖水平发生改变(P<0.001),超重或肥胖加剧上述改变(P<0.01或P<0.001)。miR-24水平与C肽水平具有相关性。结论miR-24的表达水平升高与OSAHS引起的胰岛β细胞功能紊乱相关。
Objective To investigate the expression of microRNA-24(miR-24)in the serum of patients with obstructive sleep apnea hypopnea syndrome(OSAHS)and its relationship with the impaired pancreatic β cell function.Methods 80 healthy volunteers with normal body mass index(BMI),80 OSAHS patients with normal BMI,and 80 OSAHS patients with BMI≥25 kg/m2 were recruited.All subjects underwent polysomnography.The serum level of miR-24 was detected by real-time PCR.The c-peptide level was measured by ELISA.The blood glucose level was assessed by automatic biochemical analyzer.Results As compared with normal control group,the serum level of miR-24 was up-regulated in OSAHS patients with normal BMI(P<0.001),which was further elevated in overweight or obese OSAHS patients(P<0.001).Pancreatic β-cell dysfunction was found in OSAHS patients,as evidenced by changes in c-peptide levels at 30,60,120 min after glucose stimulation and fasting and postprandial blood glucose levels(P<0.001).Overweight or obese aggravated OSAHS-induced pancreatic β-cell dysfunction(P<0.01 or P<0.001).miR-24 level was correlated with c-peptide level.Conclusion Up-regulation of serum miR-24 level was associated with OSAHS-mediated pancreatic β-cell dysfunction.
