牙周炎伴口臭患者的唾液微生物群落分析 被引量:1

摘要 目的收集牙周炎伴或不伴有口臭患者的唾液,研究其微生物群落组成的差异,探讨菌群与口臭的关系。方法于2020年9月至2021年1月就诊于郑州大学第一附属医院牙周科的患者中随机选取3例牙周炎伴口臭者(H组),3例牙周炎不伴口臭者(P组),另外选取3名健康对照者(N组)。收集他们的唾液样本,记录牙周临床指标。采用16SrRNA方法对唾液微生物群落进行从整体结构到分类学水平上组成特征的描述。结果经过对唾液样本进行分析,得到770种运算分类单位(OTUs)聚类,共同检测出丰度较高的菌群为普氏菌属、韦荣菌属、拟杆菌属、奈瑟菌属、放线菌属、嗜血杆菌属、链球菌属、梭杆菌属等。H组检测出丰度较高的菌群有梭杆菌属、奈瑟菌属、卟啉单胞菌属、坦氏菌属等,P组中丰度较高的菌群有拟杆菌属、放线菌属、链球菌属等。α多样性分析发现,H组和P组菌群多样性大于N组,H组菌群多样性大于P组(P<0.05)。β多样性分析显示,3组优势菌群有差异,N组的主要优势菌群为嗜血杆菌属。结论唾液微生物菌群的多样性和结构对口臭有影响。有无口臭的牙周炎患者的唾液微生物群落的构成差异显著。 Objective To collect the saliva of patients with periodontitis with or without halitosis,and the difference in the composition of the microbial community was studied,and the relationship between the flora and halitosis was explored.Methods From September 2020 to January 2021,3 cases of periodontitis with halitosis(group H),3 cases of periodontitis without halitosis(group P)and 3 healthy controls(group N)were randomly selected from the patients who were treated in the Periodontal Department of the First Affiliated Hospital of Zhengzhou University.Their saliva samples were collected and periodontal clinical indicators were recorded.16SrRNA method was used to describe the composition characteristics of saliva microbial community from the overall structure to the taxonomic level.Results After analyzing saliva samples,770 kinds of operational taxonomic units(OTUs)clusters were obtained,and the more abundant bacterial groups were jointly detected as Prevotella,Veronella,Bacteroides,Neisseria,Actinomyces,Haemophilus,Streptococcus and Fusobacterium.The flora with higher abundance detected in group H were Fusobacterium,Neisseria,Porphyromonas and Tanneria,while the flora with higher abundance in group P were Bacteroides,Actinomycetes and Streptococcus.The α-diversity analysis showed that the flora diversity of group H and group P was greater than that of group N,and the flora diversity of group H was greater than that of group P(P<0.05).β-diversity analysis showed that there were differences among the three groups,and Haemophilus was the main dominant group in group N.Conclusion The diversity and structure of salivary microbial flora have an effect on halitosis.There are significant differences in the composition of saliva microbial community in periodontitis patients with or without halitosis.
