摘要
甜菜是藜科两年生植物,是我国重要的耐盐糖料作物。以甜菜(Beta vulgaris L.)710-Mu品系为研究对象,利用转录组数据获得盐胁迫下35个RING型E3泛素连接酶基因,从中筛选出9个上调表达的RING型基因,用于后续研究。对这些RING基因进行了保守结构域、染色体定位、理化性质和蛋白互作网络等生物信息学分析。发现9个RING基因在Chrom02、Chrom05、Chrom06、Chrom07、Chrom08和Chrom09上均有分布,并且具有典型的RING-H2型结构域,是潜在的编码RING型泛素连接酶的基因。蛋白质互作分析结果表明,这些RING型蛋白质之间存在相互作用,并与泛素蛋白酶体系统中其他组分,如泛素藕合酶以及26S蛋白酶体,共同作用调控蛋白质稳态。荧光定量PCR结果说明,转录组数据可信,可作为下一步筛选基因的参照。这些结果可为后续深入研究甜菜泛素连接酶提供理论基础。
Sugar beet is a biennial plant of Chenopodiaceae and an important salt-tolerant sugar crop in China.Taking suger beet(Beta vulgaris L.)710-Mu strain as the research object,the transcriptome data were used to obtain 35 RING genes under salt stress,and 9 up-regulated RING genes were screened out for subsequent research.Bioinformatics analysis,such as conserved domains,chromosome location,physical and chemical properties,protein interaction network was performed.It was found that 9 RING genes were distributed on Chrom02,Chrom05,Chrom06,Chrom07,Chrom08,Chrom09,and had typical RING-H2 type domains,which were potential genes encoding RING-type ubiquitin ligase.The results of protein interaction analysis showed that these RING-type proteins interacted with other components in the ubiquitin proteasome system,such as ubiquitin lotus synthase and 26 S proteasome,to regulate protein homeostasis.The results of fluorescence quantitative PCR showed that the transcriptome data were reliable and could be used as a reference for subsequent gene screening.These results could provide a theoretical basis for the further study of beet ubiquitin ligase.
