健脾消癌方对结直肠癌干细胞增殖、迁移和侵袭的影响

Effect of Jianpi Xiaoai Recipe on the Proliferation,Migration and Invasion of Colorectal Cancer Stem Cells

目的探究健脾消癌方对结直肠癌干细胞增殖、迁移和侵袭的影响。方法从人结肠癌Lovo细胞株中获得结直肠癌干细胞,细胞表面标记物双标法流式检测鉴定结直肠癌干细胞。制备低、中、高剂量的含药血清,设立低、中、高剂量组,正常组及对照组。采用CCK-8法及Transwell实验观察健脾消癌方对结直肠癌干细胞增殖、迁移和侵袭能力的影响。结果流式细胞术结果显示微球体细胞大量表达CD133、CD44,并较贴壁细胞多。CCK-8法及Transwell实验结果发现低、中、高剂量组细胞较正常组及对照组细胞增殖、迁移及侵袭能力减弱,差异有统计学意义(P<0.05),高剂量组细胞较中、低细胞增殖能力减弱,差异有统计学意义(P<0.05),高剂量组较低剂量组迁移及侵袭能力减弱,差异有统计学意义(P<0.05)。结论健脾消癌方可抑制结直肠癌干细胞增殖、迁移和侵袭的能力,且高剂量组抑制作用明显,可通过降低癌细胞增长能力,减少癌细胞扩散抑制结肠癌的发展。

Objective To explore the effects of Jianpi Xiaoai Recipe on the proliferation,migration and invasion of colorectal cancer stem cells.Methods Colorectal cancer stem cells were obtained from the human colon cancer Lovo cell line,and the colorectal cancer stem cells were identified by double-labeling of cell surface markers.Prepare low,medium and high doses of medicated serum,and set up low,medium and high dose group,normal group and a control group.CCK-8 method and Transwell experiment were used to observe the effect of JianpiXiaoai Fang on the proliferation,migration and invasion of colorectal cancer stem cells.Results The results of flow cytometry showed that the microsphere cells expressed a large amount of CD133 and CD44 and more than adherent cells.The results of CCK-8 method and Transwell experiment found that the cells in the low,medium,and high dose groups proliferated,and their migration and invasion capabilities were weaker than those in the normal group and the control group(P<0.05),and the cells in the high dose group had weaker proliferation capabilities than the medium and low cells(P<0.05),cell migration and invasion ability of the high-dose group was weakened in the lower-dose group(P<0.05).Conclusion Jianpi Xiaoai Decoction can inhibit the proliferation,migration and invasion of colorectal cancer stem cells,and the high-dose group has a significant inhibitory effect.It can inhibit the development of colon cancer by reducing the growth of cancer cells and reducing the spread of cancer cells.
