败毒饮的高效液相指纹图谱及4种成分的含量测定 被引量:2

摘要 目的建立败毒饮的指纹图谱,并对4种成分的含量进行测定。方法采用安捷伦ZORBAX SB-C_(18)色谱柱(4.6 mm×150 mm,5 μm),以乙腈-0.1%磷酸为流动相进行梯度洗脱,检测波长334 nm,柱温30℃。通过相似度软件对11批败毒饮的指纹图谱进行质量评价,对指认的4个成分进行含量测定。与单味药进行比对,确定共有峰归属。结果建立了败毒饮的特征指纹图谱,得到16个共有峰,相似度在0.995以上,通过对照品指认出绿原酸、咖啡酸、黄芩苷、蒙花苷,其分别在19.92~199.2 ng(r^(2)=0.9994)、7.92~79.2 ng(r^(2)=0.9995)、248~2480 ng(r^(2)=0.9994)、78.16~781.6 ng(r^(2)=0.9995)与峰面积呈现良好的线性关系。结论所建立的方法简单,准确可靠,可用于败毒饮的质量控制。 Objective To establish the HPLC fingerprints of Baidu decoction and determine the content of 4 components.Methods HPLC analysis was preformed on an Agilent ZORBAX SBC18 column(4.6 mm×150 mm,5μm)with the temperature of 30℃.The mobile phase consisted of acetonitrile-0.1%phosphoric acid with gradient elution,the detection wavelength was 344 nm,and the column temperature was 30℃.Similarity was used to evaluate the fingerprint of the 11 batches of Baidu decoction,and the content of 4 components was determined simultaneously.Attribution of the common peaks was analyzed by comparing the chromatograms of single constituent drugs.Results The fingerprints of Baidu decoction showed that the similarity was greater than 0.995,and 16 common peaks were obtained.The components of chlorogenic acid,caffeic acid,baicalin,and buddleoside were identified with the reference substances.The 4 components showed good linearity at 19.92~199.2 ng(r^(2)=0.9994),7.92~79.2 ng(r^(2)=0.9995),248~2480 ng(r^(2)=0.9994),and 78.16~781.6 ng(r^(2)=0.9995).Conclusion The method is simple,accurate and reliable,which can be used for the quality control of the Baidu decoction.
