金针菇漆酶基因家族在不同农业废弃物培养基上表达情况

Expression Profiles of Laccase Gene Family in Flammulina filiformis Cultivated on Different Agricultural Waste Media

摘要 测定金针菇(Flammulina filiformis)菌丝在棉籽壳、木屑、玉米芯、玉米秸秆、柠条枝条、甘蔗渣等6种农业废弃物培养基上的生长情况、漆酶活性,并通过实时荧光定量PCR方法检测漆酶基因家族11种漆酶基因表达量。结果显示:与对照(葡萄糖培养基)比较,金针菇菌丝在甘蔗渣培养基上生长速度较慢,在其他农业废弃物培养基上生长速度较快;在玉米秸秆培养基中漆酶活性最高;在棉籽壳、柠条枝条培养基上,Lac4表达量最高;在甘蔗渣培养基上,Lac4表达量较高;在木屑、玉米芯和玉米秸秆培养基上,Lac6表达量最高。 According to the whole genome information of Flammulina filiformis,there are 11 laccase genes.They play an important role in lignin decomposition and utilization,and thus promote mycelial growth.The expression profile of the 11 genes was studied together with mycelial growth and laccase activity in F.filiformis cultivated on six kinds of agricultural wastes,including cottonseed husk,sawdust,corn cob,corn straw,Caragana korshinskii,and bagasse.The results showed that except the bagasse medium,the mycelial growth rate of F.filiformis was greater on agricultural wastes than on the glucose medium.Laccase activity was the highest on the corn straw medium(124.50 U·mL^(-1)).The expression of Lac4 was the highest on cottonseed husk and C.korshinskii,and also relatively high on bagasse.The expression of Lac6 was the highest on sawdust,corn cob and corn straw.
