高强度间歇运动对高脂饮食大鼠血液T细胞亚群及血清相关炎性因子的影响

摘要 目的:研究高强度间歇运动对高脂饮食和普通饮食大鼠血液T细胞亚群和血清相关炎性因子的影响。方法:3周龄雄性SD大鼠45只,随机分为普通饮食组(N组)和高脂饮食组(R组)。进行8周的喂养,建立肥胖模型。建模成功后分为普通饮食安静组(NC组)、高脂饮食安静组(HC组)、普通饮食运动组(NE组)、高脂饮食运动组(HE组)。高强度间歇训练8周后进行样本采集。用ELISA法检测血清IL-2、IL-6、IL-10和TNF-α的水平,用流式细胞仪检测血液CD3^(+)、CD4^(+)和CD8^(+)T细胞。结果:高脂饮食显著增加大鼠体重和TNF-α水平,饮食和运动对CD4^(+)、CD8^(+)、CD4^(+)/CD8^(+)和TNF-α有交互作用。高强度间歇运动干预下,普通饮食大鼠CD4^(+)T细胞含量和CD4^(+)/CD8^(+)的值显著增高,CD8^(+)T细胞含量显著性下降;高强度间歇运动对高脂饮食大鼠T细胞的影响则相反。结论:高强度间歇运动能够提高普通饮食大鼠的免疫机能,抑制肥胖大鼠的免疫力。高强度间歇运动和高脂饮食对大鼠血液T细胞有交互作用,可能抑制其免疫力。 Objectives:This paper studies the effects of high-intensity intermittent exercise on blood T-cell subsets and serum-related inflammatory factors in rats fed a high-fat diet and a normal diet.Methods:Forty-five 3-week-old male SD rats were randomly divided into normal diet group(N group)and high-fat diet group(R group)and fed for 8 weeks to establish an obesity model.After successful modeling,they were divided into normal diet quiet group(NC group),high-fat diet quiet group(HC group),normal diet exercise group(NE group),and high-fat diet exercise group(HE group).Samples were collected after 8 weeks of high-intensity interval training.Serum IL-2 IL-6 IL-10 and TNF-αlevels were measured by ELISA,and blood CD3^(+),CD4^(+)and CD8^(+)T cells were measured by flow cytometry.Results:High-fat diet significantly increased body weight and TNF-αlevels in rats,and diet and exercise had interactive effects on CD4^(+),CD8^(+),CD4^(+)/CD8^(+)and TNF-α.Under the intervention of high-intensity intermittent exercise,the CD4^(+)T-cell content and CD4^(+)/CD8^(+)values were significantly increased and the CD8^(+)T-cell content was significantly decreased in rats on a normal diet;the opposite effect was observed in T cell of rats on a high-fat diet with high-intensity intermittent exercise.Conclusions:High-intensity intermittent exercise can improve the immune function of normal diet rats and suppress the immunity of obese rats.High intensity intermittent exercise and high-fat diet had an interactive effect on blood T cells in rats,and this effect may suppress their immunity.
