摘要
目的探究黄连素(berberine)、葛根素(puerarin)联合应用对糖尿病大鼠血糖、血脂的影响。方法将大鼠随机分为正常对照组和造模组。造模组采用高脂高糖饲料喂养结合链脲佐菌素腹腔注射的方法建立糖尿病大鼠模型。将造模成功的糖尿病大鼠随机分为糖尿病对照组、二甲双胍组、黄连素组、葛根素组、黄连素+葛根素组。正常对照组和糖尿病对照组给予生理盐水灌胃,其余各组以相应的药物灌胃,共8周。给药期间监测大鼠的一般情况,8周后取血检测大鼠的空腹血糖(FBG)、糖耐量及甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平。结果与正常对照组比较,其余各组的FBG、糖耐量各时段的血糖值,TG,TC,LDL-C水平升高,HDL-C水平降低(P<0.01);与糖尿病对照组比较,二甲双胍组、黄连素组、葛根素组、黄连素+葛根素组的FBG、糖耐量各时段的血糖值,TG,TC,LDL-C水平降低,HDL-C水平升高(P<0.05);与黄连素组、葛根素组比较,黄连素+葛根素组的FBG、糖耐量各时段的血糖值,TG,TC,LDL-C水平降低,HDL-C水平升高(P<0.05)。结论黄连素与葛根素联合应用能降低糖尿病大鼠空腹血糖、改善糖耐量和血脂异常,其效果优于两药单独应用。
Objective To explore the effect of the combination of berberine and puerarin on blood glucose and serum lipid in diabetic rats.Methods The rats were randomly divided into a normal group and a model group.The model group rats were fed with high fat and high sugar diet combined with intraperitoneal injection of streptozotocin were used to build diabetic model rats.Diabetic rats with successful modeling were randomly divided into a diabetic control group,a metformin group,a berberine group,a puerarin group,and a berberine combined with puerarin group.The normal group and the diabetic control group were given the same volume of normal saline,and the other groups were given the corresponding drug by gavage for eight weeks.The general conditions of rats were monitored during drug administration.After eight weeks,fasting blood glucose(FBG),oral glucose tolerance test,the serum concentrations of triglyceride(TG),total cholesterol(TC),low density lipoprotein cholesterol(LDL-C),and high density lipoprotein cholesterol(HDL-C)were measured.Results Compared with the normal group,the levels of FBG,glucose tolerance,TG,TC,and LDL-C were increased and HDL-C was decreased in other groups(P<0.01).Compared with the diabetic control group,the levels of FBG,glucose tolerance,TG,TC and LDL-C in the metformin group,the berberine group,the puerarin group and the berberine combined with puerarin group were decreased,while the levels of HDL-C was increased(P<0.05).Compared with the diabetic control group,the berberine group and the puerarin group,the levels of FBG,glucose tolerance,TG,TC and LDL-C were decreased and HDL-C was increased in the berberine combined with puerarin group(P<0.05).Conclusion The combined application of berberine and puerarin could reduce FBG,improve glucose tolerance and dyslipidemia in diabetic rats,and the effect is superior to that of the two drugs alone.
