摘要
通过对甘蓝型油菜A8和A5染色体已知序列片段与显性核不育恢复基因BnMS5a和临保基因BnMS5c序列比对,确定SNP位点,并设计相关引物SEF和SER,在甘蓝型油菜显性核不育恢复系和临保系中扩增。结果表明:以恢复系DNA为模板进行扩增,出现1051 bp和924 bp两条扩增带,而以临保系DNA为模板进行扩增,只有1051 bp一条扩增带,并在测交群体3008和3012中得到验证。应用该分子标记对甘蓝型油菜显性核不育杂交种‘核杂17号’进行纯度鉴定,显示检测纯度达93.8%,准确率为100%。本研究为油菜分子育种提供了一种新标记,有助于杂交种纯度的鉴定。
By comparing the known sequence fragments of A8 and A5 chromosomes of Brassica napus with the sequences of dominant genic male sterility restorer gene BnMS5a and temporary maintainer line gene BnMS5c,the SNP loci were determined,and the related primers SEF and SER were designed to amplify in dominant genic male sterility restorer line and temporary maintainer line of B.napus.The results showed that when the restorer line DNA was used as the template for amplification,there were two amplification bands of 1051 bp and 924 bp,while when the temporary maintainer line DNA was used as the template for amplification,there was only one amplification band of 1051 bp,which was verified in the testcross populations 3008 and 3012.The molecular markers were used to identify the purity of dominant genic male sterile hybrid‘Heza 17’in B.napus.The purity was 93.8%and the accuracy was 100%.This study provides a new marker for B.napus molecular breeding and is helpful to the identification of hybrid purity.
