五子衍宗丸对生精障碍大鼠生精功能的保护作用研究

Protective Effect of Wuzi Yanzong Pills on Spermatogenic Function in Rats with Spermatogenic Disorder

目的:考察五子衍宗丸对实验性生精障碍大鼠的保护作用。方法:取90只雄性SD大鼠随机分为6组:正常对照组、模型组、克罗米芬阳性对照组及五子衍宗丸低、中、高剂量组。除正常对照组外,其他各组灌服雷公藤多苷40 mg/(kg·d)连续4 w建立生精障碍模型。从第4 w开始,各组按剂量灌胃给药,给药4 w后计算睾丸、附睾、精囊腺脏器指数,检测精子数量、活率和血清生殖激素水平,HE染色观察大鼠睾丸病理组织学改变,TUNEL染色观察睾丸细胞凋亡情况,免疫组化及qRT⁃PCR检测凋亡相关蛋白的表达。结果:生精障碍模型大鼠经高剂量五子衍宗丸治疗后,睾丸脏器指数、精子数量和活率及血清血清T水平显著升高,血清FSH、LH的水平显著降低;睾丸组织病理学损伤明显改善,并能抑制生精细胞的凋亡,促进睾丸组织中抑凋亡相关蛋白Bcl⁃2表达,抑制促凋亡相关蛋白Bax、Caspase⁃3、Caspase⁃9的表达。结论:五子衍宗丸对生精功能障碍大鼠具有保护作用,其机制可能与改善生精并抑制生精细胞凋亡相关。

Objective:To investigate the protective effect of Wuzi yanzong pills on experimental spermatogenic disorder rats.Methods:90 male SD rats were randomly divided into six groups,including normal control group,model group,clomiphene positive control group and Wuzi yanzong pills low,middle,high dose groups.Except for the normal control group,other groups were given 40 mg/(kg.d)of tripterygium glycosides for 4 weeks in succession to establish the spermatogenic disturbance model.Starting from the fourth week,each group was given the drug by gavage.After 4 weeks,viscera indexes of testis,epididymis and seminal vesicle were calculated,Sperm count,survival rate and serum reproductive hormone levels were measured,the histopathological changes of rat testis were ob-served by HE staining,the apoptosis of testicular cells was observed by TUNEL staining,the expression of apoptosis-related proteins was detected by immunohistochemistry and qRT-PCR.Results:After treatment with hiqh dose Wuzi yanzong pills,the testicular viscera in-dex,sperm count and survival rate,the level of serum T in sperm production obstacle model rats were significantly increased,the levels of FSH and LH were significantly decreased,the testicular tissue pathology damages were improved,and it could inhibit sperm cells ap-optosis,promote the expressions of anti-apoptotic proteins Bcl-2 and inhibit the expressions of pro-apoptotic proteins Bax,Caspase-3 and Caspase-9 in testicular tissue.Conclusion:Wuzi yanzong pills have a protective effect on spermatogenic dysfunction rats and its mecha-nism may be related to improving spermatogenic function and inhibiting spermatogenic cell apoptosis.
