基于网络药理学构建降糖康肾方及其作用研究

Study on the Establishment of Jiangtang Kangshen Formula based on Network Pharmacology and its Effects

目的:结合网络药理学和传统中医病机理论,构建治疗糖尿病肾病的新型中药复方——降糖康肾方,并研究其对糖尿病肾病模型大鼠的治疗作用。方法:首先从中药网络药理学数据库(TCMSP)中下载中药、成分、靶标和疾病数据,导入网络分析软件Cytoscape中构建网络,并从中筛选出能作用于糖尿病肾病疾病靶标的备选中药,然后根据中医病机理论进一步挑选药物,进而构建新型复方——降糖康肾方。通过动物实验研究降糖康肾方抗糖尿病肾病的作用,检测大鼠血糖、尿蛋白、血肌酐和尿素氮水平,HE染色评估肾组织的病理改变,检测大鼠肾脏组织SOD、CAT的活性。结果:构建的降糖康肾方由黄芪、杜仲、玉米须、黄连、丹参组成。降糖康肾方可显著降低糖尿病肾病大鼠的血糖、Cr、BUN和尿蛋白水平,并能明显改善肾脏组织中炎症细胞浸润、减轻肾脏的组织结构损害,并能增加肾脏的抗氧化能力。结论:运用网络药理学和传统中医病机理论,构建的治疗糖尿病肾病的新型复方——降糖康肾方,对糖尿病肾病的肾功能及肾脏组织结构有明显保护作用,其机制可能与增加肾脏的抗氧化能力相关。

Objective:To establish the Jiangtang kangshen formula for the treatment of diabetic nephropathy by combining network pharmacology and traditional Chinese medicine(TCM)pathogenesis theory,and to explore its therapeutic effect on diabetic nephropathy model rats.Methods:The data of TCM,ingredients,therapeutic targets and diseases were obtained from TCMSP databases and imported the network analysis software Cytoscape to build the network,and the alternative Chinese medicines which could act on the target of diabetic nephropathy were screened out.According to the pathogenesis theory of TCM,drugs were further selected,and then Jiangtang kangshen formula,a new prescription,was constructed.The anti⁃diabetic nephropathy effect of Jiangtang kangshen formula was studied through animal experiments.The levels of blood glucose,urine protein,serum creatinine and urea nitrogen in rats were detected,the pathological changes of kidney tissues were evaluated by HE staining,the activities of SOD and CAT in rat kidney tissues were detected.Results:Jiangtang kangshen formula was constituted by Astragali Radix,Eucommiae Cortex,Corn Stigma,Coptidis Rhizoma,Salviae Miltiorrhizae Radix et Rhizoma.Jiangtang kangshen formula could significantly reduce the levels of blood glucose,Cr,BUN and urine protein in diabetic nephropathy rats,and significantly improve the infiltration of inflammatory cells in the kidney tissue,reduce the damage of the kidney tissue structure,increase the antioxidant capacity of the kidney.Conclusion:Using network pharmacology and the pathogenesis theory of TCM,the new prescription for treating diabetic nephropathy,Jiangtang kangshen formula,has an obvious protective effect on renal function and renal tissue structure of diabetic nephropathy,and its mechanism may be related to increasing the antioxidant capacity of the kidney.
