摘要
采用形态学鉴定和高通量测序技术结合的方法,对松嫩盐碱草地紫花地丁根围土壤中AM真菌进行调查。形态学共鉴定出11属30种AM真菌,其中球囊霉属(Glomus)种类最多,共9种。以科学性为前提的基础上,为进一步深入研究紫花地丁根围AM真菌种类,通过高通量测序共得到7属103种虚拟种(Virtual taxa,VT):球囊霉属共82种,近明球囊霉属(Claroideoglomus)共10种,类球囊霉属(Paraglomus)共5种,多样孢囊霉属(Diversispora)共3种,盾巨孢囊霉属(Scutellospora)、无梗囊霉属(Acaulospora)、双型囊霉属(Ambispora)各1种。进一步得到了球囊霉属为松嫩盐碱草地紫花地丁根围土壤AM真菌优势属,Glomus sp.VTX00222为优势种。通过形态学鉴定和高通量测序技术相结合,弥补单种方法的局限性,提高结果的科学性和多样性。
By combining morphological identification and high-throughput sequencing,we investigated the AM fungi in the rhizosphere of Viola philippica in Songnen saline-alkali grassland.A total of 30 AM fungal species of 11 genera were identified by morphologically,including 9 species of Glomus.By exploring AM fungal diversity in the rhizosphere of V.philippica,103 virtualtaxa(VT)of 7 genera were obtained by high-throughput sequencing,including 82 VT of Glomus,10 VT of Claroideoglomus,5 VT of Paraglomus,3 VT of Diversispora,and 1 VT of Scutellospora,Acaulospora,Ambispora,respectively.Among which Glomus was the dominant genus of AM fungi in the rhizosphere of V.philippica in Songnen saline-alkali grassland,and the Glomus sp.VTX00222 was the dominant species.Through the combination of morphological identification and high-throughput sequencing technology,the limitation of single method was made up,and the scientificity and diversity of results were improved.
