Cloning and Expression of CjDFR Gene of Three Cryptotaenia japonica Varieties with Different Leaf Shape and their Correlations with Anthocyanin Content

摘要 【目的】探明不同颜色鸭儿芹中花青素的含量及其调控基因CjDFR,为进一步开发鸭儿芹中花青素及不同品种鸭儿芹的综合利用提供参考。【方法】以圆叶鸭儿芹(RL,绿色)、浅裂叶鸭儿芹(LL,绿色偏浅紫色)和深裂叶鸭儿芹(DL,绿色偏紫色)3种鸭儿芹为材料,比较分析三者花青素含量,并对其花青素合成调控基因CjDFR进行研究。【结果】花青素在DL叶片和叶柄中的含量最高,LL次之,RL中含量相对最低。经测序3种鸭儿芹材料的CjDFR长度均为1005 bp,均可编码334个氨基酸残基;3种材料的CjDFR序列同源性为99.54%,有14个位置不同,其推导的氨基酸序列同源性为98.90%,存在11个位点不同;经进化分析,鸭儿芹的CjDFR与其他伞形科的DFR处在同一分枝上,其他科植物的DFR也处在各科分枝上。DL和LL中CjDFR在叶片、叶柄中表达量显著高于RL。【结论】3种不同颜色的鸭儿芹材料花青素存在一定差异,其中花青素含量与材料颜色呈正相关,CjDFR的表达量也与鸭儿芹花青素含量呈正相关。 【Objective】The anthocyanin content and its regulatory gene CjDFR of different Cryptotaenia japonica varieties with different color are proved to provide a reference for further development of anthocyanin in Cryptotaenia japonica and comprehensive utilization of different Cryptotaenia japonica varieties.【Method】The anthocyanin content of three C.japonica varieties with round leaf(RL),lobed leaf(LL)and deeply lobed leaf(DL)is compared and the regulatory gene CjDFR of controlling anthocyanin synthesis is studied.【Result】The anthocyanin content in leaf and petiole of three C.japonica varieties is DL>LL>RL.The CjDFR sequence length of three C.japonica varieties is 1005 bp coding 334 amino acids.The CjDFR sequence homology of three C.japonica varieties reaches 99.54%with 14 different loci.The homology of the deduced amino acid sequence is 98.90%with 11 different loci.The evolution analysis shows that the CjDFR of C.japonica and the DFR of other umbelliferae both are on the same branch.The DFR of plants from other families is on the branch of each family also.The CjDFR expression in leaves and petioles of DL and LL is significantly higher than RL.【Conclusion】There is a certain difference in anthocyanin content among three C.japonica varieties.The anthocyanin content is positively related to color of C.japonica and the CjDFR expression is positively related to anthocyanin content of C.japonica also.
