摘要
目的探索和检测天冬酰胺合成酶(ASNS)在肝癌中的表达并探索其临床预后意义。方法利用TCGA肝癌公共数据,检测ASNS基因在肝癌和正常组中的差异表达,利用log-rank进行生存分析并绘制kaplan-meier曲线,同时采用单因素和多因素cox分析ASNS基因在肝癌预后的预测价值并绘制列线图,并进一步采用免疫组化检测我院20例肝癌术后样本中的ASNS蛋白表达情况。结果与正常组相比,ASNS基因和蛋白在肝癌中均呈明显高表达(P<0.05),且高表达ASNS的肝癌患者生存时间显著短于其低表达组,单因素和多因素cox分析均显示,ASNS的表达水平可以作为肝癌患者术后生存率的独立预测因子。结论 ASNS在肝癌中呈高表达,且其表达水平可以作为肝癌患者术后生存率的独立预测因子,值得进一步深入研究。
Objective To explore and detect the expression of asparagine synthetase(ASNS) in hepatocellular carcinoma and explore its clinical prognostic significance. Methods Using TCGA hepatocellular carcinoma public data, we detected the differential expression of ASNS gene in hepatocellular carcinoma and normal group, and used log-rank for survival analysis and plotted kaplan-meier curve, and also used univariate and multifactor cox to analyze the predictive value of ASNS gene in hepatocellular carcinoma prognosis and plotted column line graph, and the ASNS protein expression in the postoperative samples of 20 cases of hepatocellular carcinoma in our hospital was further detected by immunohistochemistry. Results Compared with the normal group, both ASNS gene and protein showed significantly higher expression in hepatocellular carcinoma(P<0.05), and the survival time of hepatocellular carcinoma patients with high ASNS expression was significantly shorter than that of their low expression group. Both univariate and multifactorial cox analyses showed that the expression level of ASNS could be an independent predictor of postoperative survival of hepatocellular carcinoma patients. Conclusions ASNS is highly expressed in hepatocellular carcinoma, and its expression level can be an independent predictor of postoperative survival of hepatocellular carcinoma patients, which deserves further in-depth study.
