长链非编码RNA AFAP1-AS1在卵巢癌中的表达及预后意义

Expression and prognostic significance of long non-coding RNA AFAP1-AS1 in ovarian cancer

目的分析卵巢癌患者长链非编码RNA(lncRNA)肌动蛋白纤维相关蛋白1-反义RNA1(AFAP1-AS1表达水平,并探讨其与预后的关系。方法收集卵巢癌组织96例及正常卵巢组织36例为研究对象。采用实时荧光定量PCR法检测组织AFAP1-AS1表达水平。分析AFAP1-AS1与患者临床病理特征的关系,Kaplan-Meier分析AFAP1-AS1表达情况与卵巢癌患者生存情况的关系,采用COX法分析影响卵巢癌患者预后的因素。结果与正常卵巢组织相比,卵巢癌组织AFAP1-AS1相对表达水平较高(P<0.05)。AFAP1-AS1与淋巴结转移、按照国际妇产科联盟(FIGO)分期、病理分级相关(P<0.05)。Kaplan-Meier法显示AFAP1-AS1低表达患者平均生存时间为(32.33±0.80)个月,显著长于AFAP1-AS1高表达卵巢癌患者[(23.19±1.45)个月](P<0.05)。多因素分析表明,FIGO分期(Ⅲ~Ⅳ期)、病理分级(低、中分化)、淋巴结有转移、AFAP1-AS1高表达是影响卵巢癌患者不良预后的独立危险因素(P<0.05)。结论 lncRNA AFAP1-AS1在卵巢癌组织中表达上调,且与患者术后不良预后密切相关。

Objective To analyze the expression level of long non-coding RNA(lncRNA) actin filament-associated protein1-anti-sense RNA1(AFAP1-AS1) in patients with ovarian cancer,and to explore its relationship with prognosis.Methods A total of 96 cases of ovarian cancer tissue and 36 cases of normal ovarian tissue were selected as research objects.The expression level of AFAP1-AS1 was detected by real-time fluorescent quantitative PCR.The relationship between AFAP1-AS1 and clinicopathological characteristics of patients was analysised.Kaplan-Meier was used to analyze the relationship between the expression of AFAP1-AS1 and the survival of patients with ovarian cancer,and COX method was used to analyze the prognostic factors of patients with ovarian cancer.Results Compared with that in normal ovarian tissue,the relative expression level of AFAP1-AS1 in ovarian cancer was higher(P<0.05).AFAP1-AS1 was correlated with lymph node metastasis,International Federation of Gynecology and Obstetrics(FIGO) stage and pathological grade(P<0.05).Kaplan-Meier method showed that the mean survival time of patients with low expression of AFAP1-AS1 was(32.33±0.80)months,which was significantly longer than that of patients with high expression of AFAP1-AS1 [(23.19±1.45) months](P<0.05).Multivariate analysis showed that FIGO stage(Ⅲ~Ⅳ),pathological grade(low and middle differentiation),metastasis of lymph nodes and high expression of AFAP1-AS1 were independent risk factors for poor prognosis of patients with ovarian cancer(P<0.05).Conclusion The expression of lncRNA AFAP1-AS1 is up-regulated in ovarian cancer,which is closely related to the poor prognosis.
