芪枝渴痹通方对糖尿病小鼠行为学与坐骨神经病理学的影响 被引量:3

Effects of Qizhike Bitong Formula on behavior and sciatic nerve pathology in diabetic mice

摘要 目的:观察芪枝渴痹通方(QZKBTF)对STZ诱导的糖尿病小鼠行为学与坐骨神经病理学的影响。方法:C57BL小鼠单次腹腔注射120mg/kg STZ建立糖尿病模型,造模成功后随机分为模型组,QZKBTF低、中、高剂量组与甲钴胺组,各10只,设立正常组(n=10),并进行12周的药物干预。使用智能热板仪测定热缩足反射潜伏期(TWL),恒温水浴锅测定甩尾潜伏期(TFL),PowerLab8/35八通道生理记录仪测定运动神经传导速度(MNCV)及感觉神经传导速度(SNCV),取小鼠坐骨神经进行病理学检测。结果:在8周、12周,与正常组比较,模型组小鼠TWL、TFL均显著降低(P<0.01),QZKBTF各剂量组和甲钴胺组小鼠TWL、TFL较模型组均显著增加(P<0.01,P<0.05)。在12周,与正常组比较,模型组小鼠MNCV与SNCV均显著减慢(P<0.01),QZKBTF各剂量组和甲钴胺组小鼠MNCV与SNCV较模型组显著提升(P<0.01,P<0.05)。QZKBTF和甲钴胺可改善糖尿病小鼠坐骨神经纤维排列,髓鞘板层结构、轴突的病理改变。结论:QZKBTF可缓解糖尿病小鼠痛温觉过敏,提高神经传导速度,改善坐骨神经病理结构。 Objective: To observe the effects of Qizhike Bitong Formula(QZKBTF) on behavior and sciatic nerve pathology of STZ-induced diabetic mice. Methods: C57 BL mice were injected intraperitoneally with 120 mg/kg STZ to establish diabetic model. After successful model establishment, the mice were randomly divided into a model group, low-, medium-and high-dose QZKBTF group and mecobalamin group(n=10). The normal group(n=10) was established, and drug intervention was carried out for 12 weeks. The reflex latency(TWL) of heat-shrinkable foo was measured with intelligent hot plate instrument, tail flick latency(TFL) was measured with constant temperature water bath, motor nerve conduction velocity(MNCV) and sensory nerve conduction velocity(SNCV) were measured with PowerLab8/35 eight-channel physiological recorder, and sciatic nerve of mice was taken for pathological examination. Results: At the 8 th and 12 th week, compared with the normal group, the TWL and TFL in the model group were significantly decreased(P<0.01), while the TWL and TFL in QZKBTF and mecobalamin groups were increased(P<0.01, P<0.05). At the 12 th week, compared with the normal group, MNCV and SNCV in the model group were significantly slowed down(P<0.01), while MNCV and SNCV in QZKBTF and mecobalamin groups were increased(P<0.01, P<0.05). QZKBTF and mecobalamin could improve the arrangement of sciatic nerve fibers,the lamina structure of myelin sheath and the pathological changes of axons in diabetic mice. Conclusion: QZKBTF can relieve pain and temperature hypersensitivity, increase nerve conduction velocity and improve sciatic nerve pathological structure in diabetic mice.
