钙释放激活钙通道蛋白1和基质相互作用分子1对哮喘大鼠气管收缩的影响 被引量:5

Effect of Calcium release-activated calcium channel protein 1 and stromal interacting molecular 1 on airway contraction in asthmatic rats

摘要 目的研究钙释放激活钙通道蛋白1 (Orai1)和基质相互作用分子1(STIM1)对哮喘大鼠气管收缩的影响。方法将实验动物随机分为正常组与模型组,每组15只。正常组于第1,8天腹腔注射灭菌磷酸盐缓冲溶液(PBS),于第15~20天雾化吸入灭菌PBS,每天1次;模型组于第1,8天腹腔注射新鲜的卵清蛋白致敏液,并于第15~20天雾化吸入1%卵清蛋白,每天1次。用气管张力实验观察气管平滑肌钙库操纵的钙内流(SOCE)引起的气管收缩的改变,用蛋白质印迹法检测Orai1和STIM1蛋白表达水平的变化。结果造模成功后,正常组和模型组SOCE引起的气管收缩在高钾引起的收缩中占比分别为(64.16±30.74)%和(140.92±110.65)%,Orai1蛋白相对表达量分别为(42.00±20.00)%和(81.00±7.00)%,STIM1蛋白相对表达量分别为(66.00±6.00)%和(138.00±12.00)%,差异均有统计学意义(均P <0.05)。结论哮喘大鼠SOCE引起的气管收缩可能随着Orai1和STIM1蛋白表达的增高而增强。 Objective To study the effect of calcium release-activated calcium channel protein 1(Orail) and stromal interacting molecular 1(STIM1) on tracheal contraction in asthmatic rats.Methods The experimental animals were randomly divided into normal group and model group with 15 rats per group.The normal group was intraperitoneally injected with sterilized phosphate buffer solution(PBS) on days 1 and 8,and atomized inhaled with sterilized PBS on days 15 to 20,once a day.The model group was intraperitoneally injected with fresh ovalbumin sensitization solution on days 1 and 8,and atomized with 1 % ovalbumin inhalation on days 15 to 20,once a day.Tracheal tension test was used to observe the changes of tracheal contraction induced by store-operated Ca^(2+)entry(SOCE).Western blot was used to detect the changes of Orail and STIM1 protein expression.Results After obtained successful animal model,the proportion of SOCE-induced tracheal contraction in high potassium-induced tracheal contraction in normal group and model group was(64.16±30.74) % and(140.92±110.65)%,the relative expression levels of Orail protein were(42.00±20.00) % and(81.00±7.00) %,the relative expression levels of STIM1 protein in the normal group and the model group were(66.00±6.00) % and(138.00±12.00) %.The differences were statistically significant(all P <0.05).Conclusion The tracheal contraction induced by SOCE in asthmatic rats may be enhanced with the increased expression of Orail and STIM1 proteins.
