2015-2020年我国禽腺病毒流行病学调查及高致病性FAdV-4分离株遗传变异性分析 被引量:14

Epidemiological Survey of Avian Adenovirus in China from 2015 to 2020 and Analysis of Genetic Variability of Highly Pathogenic FAdV-4 Isolates

摘要 为了解目前我国Ⅰ群禽腺病毒的流行情况,本中心以临床诊断、分子生物学检测、血清学检测和LMH细胞病毒分离等方法对全国重点养殖家禽和水禽的区域进行跟踪检测。本实验室于2015-2020年,从北京、天津、河北、山东等30余个省份共收集到7098份疑似样品,hexon基因测序成功1668份,其中FAdV-1型129份、FAdV-2型89份、FAdV-3型34份、FAdV-4型638份、FAdV-5型36份、FAdV-6型5份、FAdV-7型13份、FAdV-8a型177份、FAdV-8b型193份、FAdV-9型8份、FAdV-10型135份和FAdV-11型211份。将FAdV阴性样品全部接种LMH细胞进行病毒分离,共得到分离株703株,其中FAdV-4型分离株439株。高致病性FAdV-4型437株,与高致病性FAdV-4参考株同源性为99.1%~100%,非致病性分离株2株,与ON1参考株同源性为99.6%~100%。hexon基因的188、193、195、238和240位氨基酸也出现变异。 In order to understand the prevalence of Poultry adenovirus in China,the center used methods such as clinical diagnosis,molecular biological testing,serological testing and LMH cell virus isolation to track and test key poultry and waterfowl breeding areas across the country.From 2015 to 2020,our laboratory collected 7098 suspected samples from more than 30 provinces including Beijing,Tianjin,Hebei,and Shandong and successful hexon gene sequencing was conducted for 1668 copies,among them,129 copies of FAdV-1,89 FAdV-2,34 FAdV-3,638 FAdV-4,36 FAdV-5,5 FAdV-6,13 FAdV-7,177 copies of FAdV-8a,193 copies of FAdV-8b,8 copies of FAdV-9,135 copies of FAdV-10 and 211 copies of FAdV-11.Total 703 FAdV strains were isolated from FAdV suspected samples by seeding LMH cells.Among 439 FAdV-4 strains identifi ed,437 isolates were highly pathogenic and the amino acid homology with highly pathogenic FAdV-4 reference strains was 99.1%-100%.Other 2 isolates were non-pathogenic and the amino acid homology with ON1 reference strain was 99.6%-100%.The amino acids at 188,193,195,238 and 240 of the hexon gene also had mutations.
