摘要
阳离子脂质体是基因转染中最受关注的一种非病毒类载体。本文以双十二烷基二甲基溴化铵(DDAB)与聚山梨酯-80(Tween 80)复配构建阳离子脂质体,并评价其使用安全性和DNA压缩性能。结果显示,DDAB与Tween 805∶5复配形成的脂质体,粒径和Zeta电位适中,对溴酚蓝产生较强的包封作用;同时,可以有效地抑制DDAB的细胞毒性,在≤800μM时,红细胞溶血率<5%,表现出优良的使用安全性;当该脂质体与DNA质量比为4时,对DNA显示出较强地结合压缩作用,且形成的复合物带有较高的正电荷(约+40 mV),这为基因转染提供了良好的理论基础。
Cationic liposome is one of the most crucial non-viral vectors in gene transfection.In this study,a liposome was constructed by the combination of didecyl dimethyl ammonium bromide(DDAB)and polysorbate-80(Tween 80),and its biosafety and DNA compression performance were evaluated.The results showed that the liposome was constructed by mixing DDAB and Tween 80 with the ratio of 5∶5 possessed moderate particle size and zeta potential,which could produce strong entrapment effect on bromophenols.Moreover,at concentration less than 800μM,the liposome possessed excellent biosafety level by having less than 5%hemolysis rate and effectively inhibited the cytotoxicity of DDAB.When the mass ratio of the liposome to DNA was 4,it produced strong compression on DNA and the complex formed had a relatively high positive charge(about+40 mV)which provided a good theoretical basis for gene transfection.
