摘要
目的基于当前RNA修饰对细胞生物学功能具有多种调控作用的研究日益受到广泛关注,该文章总结了RNA的5-methlcytosine(m5C)修饰与RNA甲基转移酶NSUN2的相关研究进展。方法查阅并引用近10年来国内外发表的53篇相关文献,从m5C修饰的背景知识、检测方法、NSUN2蛋白的功能、以及与疾病的关联性等方面,将RNA的m5C修饰与NSUN2相关研究进展进行总结归纳。结果与结论NSUN2具有tRNA、mRNA、非编码RNA(ncRNA)m5C修饰活性以及非甲基转移酶活性,与肿瘤和神经系统疾病发生具有一定关联性。靶向NSUN2在疾病治疗中具有潜在的应用价值。
Objective RNA epigenetics is evolving rapidly and transforming many aspects of life science.We have summarized the recent research progress on a powerful but poorly understood RNA modification 5-methylcytosine(m5C) and its RNA methyltransferase NSUN2.Methods 53 literatures published in recent 10 years have collectively shown that m5C is detectable in multiple RNA biotypes, dynamic, conserved, functionally important, and regulates the metabolism of the modified RNAs.Results and Conclusion and conclusions NSUN2 posesses not only methyltransferase activity toward tRNA,mRNA,noncoding RNA but also non methyltransferase activity;both the two activities are related to cancer, cardiovascular and neurological diseases.Thus targeting NSUN2 may have potential application value in disease treatment.
