癌变激酶信号的重塑调控——细菌效应蛋白的新应用

摘要 细菌进化了大量效应蛋白以便利其在宿主环境中生存以及增殖。在持续的细菌—宿主相互作用过程中,这些效应蛋白进化出高度的底物特异性以及独特的催化机理,使得细菌效应蛋白在探究复杂的细胞信号调控网络以及疾病治疗中具有独特的优势。随着近期蛋白质化学与蛋白质工程领域的发展,获得了精密控制蛋白活性的手段,进一步拓展了这些细菌效应蛋白的应用潜力。本研究旨在论述利用化学生物学工具精密调控细菌效应蛋白活性,从而探究其在调控细胞癌变激酶信号中的应用。 Bacteria have evolved a large number of effector proteins to facilitate their survival and prolifera-tion in the host environment.Continuous bacteria-host rivalry has granted these effector proteins with high substrate specificities and unique catalytic mechanisms,making them exceptionally advantageous in exploring complex signal regulation networks and disease treatment.With the recent advancements in protein chemistry and biomolecular engineering,we have acquired activity control approaches for these effectors with great pre-cisions,further expanding the applications of these bacterial effector proteins.Herein,we review the utiliza-tion of chemical biology tools on bacterial effectors to reprogram these enzymes to rewire carcinogenic kinase signaling for cancer therapy.
