摘要 目的:确定各指标权重系数,优选陈皮麻黄饮水提工艺,为其工业化生产提供参考。方法:高效液相色谱法(HPLC)同时测定苦杏仁苷、橙皮苷、迷迭香酸、甘草酸含量,紫外分光光度法(UV)测定总黄酮含量;以苦杏仁苷、橙皮苷、迷迭香酸、甘草酸及总黄酮含量为综合评价指标,采用层次分析法(AHP)确定各指标权重系数,结合Box-Behnken响应面法优选陈皮麻黄饮的水提工艺,并进行验证试验。结果:苦杏仁苷、橙皮苷、迷迭香酸、甘草酸分别在0.1324~1.3240、0.0656~0.6560、0.0044~0.0441、0.0187~0.1868 mg/mL内呈良好的线性关系,加样回收率为95.32%~96.64%,RSD均小于5%;橙皮苷、苦杏仁苷、迷迭香酸、甘草酸、总黄酮含量权重系数分别为0.4498、0.1937、0.1937、0.1050、0.0577;最优水提工艺为溶媒用量12倍,提取次数3次,每次提取时间1 h。在验证试验中,预测综合得分102.61,实际综合得分(101.28±3.10),RSD为3.06%,与预测值差异较小,且在合理范围内。结论:该方法稳定可行,预测性良好,可用于优化陈皮麻黄饮的水提工艺。 Objective:To determine the weight coefficient of each index,and to optimize the water extraction process of Chenpi Mahuang Yin,so as to provide a reference for its industrial production.Method:Simultaneous determination of amygdalin,hesperidin,rosmarinic acid and glycyrrhizic acid by high performance liquid chromatography(HPLC),and determination of total flavonoids by ultraviolet spectrophotometry(UV).Taking the contents of amygdalin,hesperidin,rosmarinic acid,glycyrrhizic acid and total flavonoids as comprehensive evaluation indexes,the weight coefficients of each index were determined by analytic hierarchy process(AHP),and the water extraction process of Chenpi Mahuang Yin was optimized by box Behnken response surface method.Results:Amygdalin,hesperidin,rosmarinic acid and glycyrrhizic acid showed a good linear relationship within 0.1324-1.3240,0.0656-0.6560,0.0044-0.0441 and 0.0187-0.1868 mg/mL,respectively.The recoveries were 95.32%-96.64%,and the RSD was less than 5%.The weight coefficients of hesperidin,amygdalin,rosmarinic acid,glycyrrhizic acid and total flavonoids were 0.4498,0.1937,0.1937,0.1050,0.0577,respectively.The optimal water extraction process was 12 times the amount of solvent,3 extraction times,and each extraction time was 1 h.In the verification test,the predicted comprehensive score was 102.61,the actual comprehensive score was(101.28±3.10),and the RSD was 3.06%,which was less different from the predicted value and within a reasonable range.Conclusion:The method is stable and feasible,with good predictability,and can be used to optimize the water extraction process of Chenpi Mahuang Yin.
