摘要
建立七清败毒颗粒中黄芩苷和大黄素的含量测定方法。色谱条件:色谱柱为Kromasil 100-5-C18(4.6 mm×250 mm,5μm);流动相为甲醇(A)-0.1磷酸溶液(B);梯度洗脱程序(0~5 min,20%A;5.1~15.0 min,40%~60%A;15.1~25.0 min,60%~90%A;25.1~26.0 min,90%~60%A;26.1~30.0 min,20%A);检测波长为0~20 min 276 nm,20 min后288 nm;流速1.0 mL/min;柱温30℃;进样体积10μL。黄芩苷在2.4~123.3μg/mL范围内线性关系良好;黄芩苷的平均回收率为100.2%(RSD=1.8%);大黄素在0.3~15.0μg/mL范围内线性关系良好,大黄素的平均回收率为101.4%(RSD=0.8%)。说明所建立的方法专属性强,操作方便,结果准确,可用于七清败毒颗粒中黄芩和大黄的质量控制。
To establish a method for the determination of baicalin and emodin in Qiqing Baidu granules. Chromatographic conditions were as follows:Kromasil 100-5-C18(4.6 mm×250 mm,5 μm);mobile phase:methanol(A)-0.1% phosphoric acid solution(B);gradient elution program:0~5 min,20% A;5.1~15.0 min,40%~60% A;15.1~25.0 min,60%~90% A;25.1~26.0 min,90%~60% A;26.1~30.0 min,20% A;detection wavelength:0~20 min,276 nm,20 min later,288 nm;flow rate:1.0 mL·min^(-1);column temperature:30℃;injection volume:10 μL. The linear range of baicalin was 2.4~123.3 μg·mL^(-1).The average recovery of baicalin was 100.2%(RSD=1.8%). Emodin had a good linear relationship in the range of 0.3~15.0 μg·mL^(-1),the average recovery of emodin was 101.4%(RSD=0.8%). The established method had strong specificity,convenient operation,and accurate results,which can be used for the quality control of Scutellaria baicalensis and Rheum officinale in Qiqing Baidu granules.
