摘要
目的探讨Crk SH3域结合鸟嘌呤核苷酸交换因子(C3G)在胃癌组织中的表达及其临床意义。方法利用Oncomine等生物信息大数据库,分析C3G在肿瘤组织及癌旁组织中是否存在表达差异。2012年12月~2015年12月手术切除的胃癌病人121例,使用免疫组化法检测癌组织及癌旁组织中C3G蛋白的表达,分析C3G蛋白的表达与临床病理特征及预后的关系。结果C3G在胃癌组织中表达高于正常组织(P<0.05),癌组织及癌旁组织中免疫组化阳性表达率分别为70%、31%。Kaplan-Meier生存曲线分析表明,胃癌病人C3G阳性表达与其术后生存率具有相关性(P<0.05),且C3G的表达与胃癌肿瘤T分期、淋巴结的转移有关(P<0.05),与病人年龄、性别、肿瘤的分化程度无关(P>0.05)。结论C3G在胃癌组织中呈高表达,其高表达与胃癌发展及不良预后有关。
Objective To explore the expression and significance of the Crk SH3-domain-binding guanine-nucleotide exchange factor(C3 G)in the gastric cancer tissue.Methods Using Oncomine to analyze the differences in expression of C3 G in tumor tissues and para-carinoma tissues.Collecting 121 postoperation gastric cancer patients’clinical data who is up to standard,from December 2012 to December 2015.And detecting the expression of C3 G protein in their cancerous tissues and para-carinoma tissues by immunohistochemistry.Then analyzing the relationship between C3 G protein expression and clinicopathological characteristics and prognosis in these patients.Results Bioinformatics analysis results showed C3 G was high expression in gastric cancer tissue than normal tissue(P<0.05),and in immunohischemistry,the positive expression rate in cancerous tissues and para-carinoma tissues was 70%and 31%.Through Kaplan-Meier survival curve analysis,the survival rate of the patients,who the C3 G expression whether positive,was significative(χ2=4.384,P<0.05).And the expression of C3 G was related to the T stage of the tumor and lymph nodes metastasis(P<0.05),but was not to the age,sex and differentiated degree of tumor(P>0.05).Conclusion C3 G is highly expressed in gastric cancer tissues,and its high expression is related to the development of gastric cancer and its poor prognosis.
