沙柳SpsNAC005基因克隆及生物信息学分析

摘要 本研究以沙柳为材料,基于毛果杨、欧洲红皮柳的序列,采用同源克隆的方法获得沙柳SpsNAC005基因,利用生物信息学软件分析SpsNAC005基因序列、构建进化树、预测蛋白结构。结果显示,SpsNAC005基因的开放阅读框序列长度为927 bp,编码为308个氨基酸。生物信息学分析表明,SpsNAC005含有NAC基因家族的典型NAM结构域,具有A、B、C、D、E 5个保守亚结构域。SpsNAC005基因为亲水性蛋白,预测细胞定位在细胞核上,其可能作为转录激活因子在细胞核中行使功能。本研究为进一步探索NAC基因在沙柳抗逆胁迫调控中的机制研究提供了重要的科学依据。 In this study,the Salix psammophila was used as the material,and based on the sequences of Populus trichocarpa and Salix purpurea,the method of homologous cloning was used to obtain the SpsNAC005 gene of the Salix psammophila.The bioinformatic software was used to analyze the SpsNAC005 gene sequence,the phylogenetic tree was constructed,and the protein structure was predicted.The results showed that the open reading frame sequence of the SpsNAC005 gene was 927 bp in length,and encoded as 308 amino acids.The bioinformatic analysis indicated that the SpsNAC005 contained a typical NAM domain of the NAC gene family and hasd five conserved subdomains,i.e.,A,B,C,D,and E.The SpsNAC005 gene was a hydrophilic protein,which was predicted to be located on the nucleus,and it may function as a transcriptional activator in the nucleus.This paper provided an important scientific basis for further exploring the mechanism of the NAC gene in the regulation of the anti-reverse stress in the Salix psammophila.
