摘要
目的:观察当归芍药散预处理对缺血/再灌注损伤模型小鼠离体子宫生理特性的影响并分析其可能机制。方法:将60只雌性小鼠随机分为空白对照组,模型组,中药高、中、低剂量组,每组12只。各组给予对应药物处理,预处理后建立子宫缺血/再灌注损伤动物模型。成模后2 h取小鼠右侧离体子宫平滑肌条检测子宫肌条钙离子(Ca^(2+))浓度、超氧化物歧化酶(superoxide dismutase,SOD)、乳酸脱氢酶(lactate dehydrogenase,LDH)、丙二醛(malondialdehyde,MDA)及线粒体呼吸链酶复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ水平,记录5组小鼠离体子宫平滑肌条生理特性的改变情况。结果:与模型组比较,中药中、高剂量组收缩力、频率、子宫活动力、LDH、MDA、线粒体呼吸链酶复合体Ⅰ、Ⅱ、Ⅲ、Ⅳ及Ca^(2+)浓度均明显降低,并恢复到空白对照组水平,SOD则明显高于模型组和中药低剂量组(P<0.05),且这种效应存在剂量依赖性,中药高、中剂量组组间比较,差异有统计学意义(P<0.05)。结论:当归芍药散可能通过抑制线粒体呼吸链酶这一细胞能量代谢途径,阻碍Ca^(2+)内流而降低离体子宫平滑肌对缩宫素的敏感性,这会使子宫活动力降低,在一定程度上促进了缺血/再灌注子宫生理功能的恢复。
Objective:To observe the impacts of Danggui Shaoyao powder preconditioning on physiological characteristics of isolated uterus in mice with ischemia/reperfusion injury,and analyze its possible mechanism.Methods:Sixty female mice were randomized into blank control group,the model group,high,moderate and low dosages groups of herbs,12 mice in each group.Different groups were given the corresponding medicine,after preconditioning to establish the models of uterine ischemia/reperfusion injury.Smooth muscle strips of isolated right mice uterus were taken out two hours after modeling to measure the concentrations of Ca^(2+) in myometrium,the levels of SOD,LDH,MDA,mitochondrial respiratory chain enzyme complexⅠ,Ⅱ,ⅢandⅣ,to record the changes of physiological characteristics of isolated uterine smooth muscle strip of the mice in five groups.Results:Compared with the model group,contractility,frequency,uterine activity,LDH,MDA,the concentrations of mitochondrial respiratory chain enzyme complexⅠ,Ⅱ,ⅢandⅣ,and Ca^(2+) of the moderate and high doses groups of herbs lowered notably,and these restored to the levels of blank control group,the levels of SOD were higher than these of the model group and low dose group of herbs(P<0.05),and the effect was dose-dependent,the difference was statistically significant between high and moderate doses groups of herbs(P<0.05).Conclusion:Danggui Shaoyao powder could reduce the sensitivity of isolated uterine smooth muscle to oxytocin which might be through inhibiting mitochondrial respiratory chain enzyme,which is the energy metabolism pathway of cells,and blocking Ca^(2+) influx,it would reduce the activity of uterus and promote the recovery of physiological function of ischemia/reperfusion uterus to a certain extent.
