Application of DNA Barcoding to Identify Uncommon Flies in Ningbo Port

摘要 本研究对宁波口岸蝇类监测中捕获的5只本底非常见蝇进行种类鉴定。经COⅠ基因扩增及测序,将序列信息在BOLD及GenBank数据库中检索,并与其近源序列比对,计算遗传距离,建立进化树。5只蝇中,有4只经过序列分析结合形态学特征鉴定到了具体种类,有1只COⅠ基因序列在数据库中无高质量匹配,未成功鉴定。研究表明,DNA条形码技术结合形态学鉴定能够快速高效地鉴定未知种的蝇类。 Species identification was conducted for five local uncommon flies captured in the fly monitoring at Ningbo port.Subjected to COⅠgene amplification and sequencing,the sequences was aligned in BOLD and GenBank databases,and compared with their similar sequences to calculate the genetic distance and build a phylogenetic tree.Four of the five flies were identified to specific species by sequence analysis combined with morphological features,and one COⅠgene sequence was not successfully identified without high-quality matches in the database.The study shows that DNA barcoding technique combined with morphological identification can identify flies of unknown species quickly and efficiently.
