摘要
本研究采用高效液相色谱法,通过优选合适的梯度洗脱条件实现物质分离,试验以甲醇和0.5%的乙酸作为流动相,流速为0.8 mL·min-1,检测波长为290 nm,左右柱温均为30℃,建立了马铃薯酒的高效液相色谱法指纹图谱,并通过相似度分析考查了酒样的质量稳定性。随机选择10批马铃薯酒样品建立其指纹图谱,从图谱中找到5个色谱峰作为共有峰,并进行相似度分析。结果表明,相似度在0.93以上,说明10个批次样品的质量比较稳定。该法方便有效,在马铃薯酒质量控制和检验掺伪方面具有研究和实践意义。
In this study,high-performance liquid chromatography was used and the separation was achieved by optimizing the appropriate gradient elution conditions.Methanol and 0.5%acetic acid were used as mobile phase.The flow rate is 0.8 ml·min^(-1).The detection wavelength is 290 nm.The left and right column temperatures are 30℃.According to the above conditions,the high-performance liquid chromatography fingerprints of potato wine were established,and the stability of the wine was investigated by similarity analysis.The fingerprints of 10 samples of potato wine were randomly selected to establish,and 5 peaks were found as common peaks.The results showed that the similarity of the 10 batches of potato wine was more than 0.93,which indicated that the quality of the wine was stable.The results showed that the method was convenient and effective,and had research and practical significance in quality control and adulteration detection of potato wine.
