针刺对急性脑出血大鼠Nrf2/ARE信号通路关键因子表达的影响 被引量:5

Effect of Acupuncture on Key Factors Expressions of Nrf2/ARE Signaling Pathway in Rats with Acute ICH

摘要 目的:探讨“百会”透“曲鬓”针刺法对急性脑出血大鼠Nrf2/ARE信号通路关键因子表达的影响。方法:将健康雄性Wistar大鼠54只随机分为针刺组、模型组和假手术组;再将各组分成3个时间亚组分别为1 d、3 d和7 d,每个亚组6只大鼠。采用自体血注入法建立脑出血大鼠模型。针刺组于造模成功后给予大鼠患侧“百会”透“曲鬓”针刺治疗。采用Longa评分法综合评价各组大鼠的神经功能缺损情况。Western-blot方法检测大鼠脑组织血肿周围转录调节因子2(Nrf2)、血红素加氧酶1(HO-1)和醌氧化还原酶1(NQO1)蛋白表达情况。结果:神经行为学评分结果:模型组大鼠出现不同程度的神经功能缺损症状,术后3 d神经功能缺损最重,术后7 d减轻。针刺组大鼠在各个时间点的评分均低于同时间点模型组,与模型组比较,差异具有统计学意义(P<0.05)。Western blot方法检测结果:模型组大鼠Nrf2、HO-1和NQO1蛋白表达量于术后1 d已有较明显增多,术后3 d表达最多,术后7 d较高峰期有所回落,但表达仍高于假手术组,差异具有统计学意义(P<0.01)。针刺组大鼠各个时间点Nrf2、HO-1和NQO1蛋白表达量与同期模型组比较,均明显高于模型组,差异具有统计学意义(P<0.01)。各组大鼠脑组织中Nrf2与HO-1、Nrf2与NQO1表达呈正相关。结论:“百会”透“曲鬓”头针疗法可以改善大鼠神经缺损症状,促进Nrf2/ARE信号通路关键因子Nrf2、HO-1和NQO1的表达,对抗氧化应激损伤,从而保护脑细胞。 Objective:To explore the effect of needling Baihui(GV 20)through Qubin(GB 7)on key factors expressions of Nrf2/ARE signaling pathway in rats with acute intracerebral hemorrhage(ICH).Methods:54 healthy male Wistar rats were randomly divided into the acupuncture group,the model group and the sham-operation group.Each group was subdivided into three subgroups according to the time of 1 d,3 d and 7 d,with 6 rats in each subgroup.ICH model was established by autologous blood injection in the rats.After the successful modeling,the acupuncture group was treated with needling GV 20 through GB 7 on the affected side of the rats,and the samples were taken at the corresponding time points.Longa score was used to evaluate the neurological deficits of the rats in each group.Western-blot method was used to detect the expressions of Nrf2,HO-1 and NQO1 around the hematoma of rat brain tissues at different time points,and the correlation of these indexes was analyzed.Results:In terms of neurobehavioral scoring results,the rats in the model group had neurological deficits of varying degrees,the neurological deficits were most severe on the 3rd day after the operation and relieved on the 7th day;the scores in the acupuncture group were lower than those in the model group at each time point(P<0.05).Western blot results showed that the expressions of Nrf2,HO-1 and NQO1 increased significantly in the model group on the 1st day after operation,the highest expressions were on the 3rd day after operation and the expressions dropped again on the 7th day,but the expressions were still higher than those in the sham-operation group(P<0.01).The expressions of Nrf2,HO-1 and NQO1 were significantly higher in the acupuncture group than those in the model group at each time point(P<0.01).The expression of Nrf2 was positively correlated with the expressions of HO-1,NRF2 and NQO1 in brain tissues of the rats in each group.Conclusion:Scalp needling GV 20 through GB 7 can improve the symptoms of nerve defect,promote the expressions of Nrf2,HO-1 and NQO1,and resist oxidative stress injury in the rats with acute ICH,thus to protect brain cells.
