基于网络药理学研究十全育真汤治疗非小细胞肺癌的作用机制 被引量:2

Action mechanism of Shiquan Yuzhen Tang(十全育真汤)in the treatment of non-small cell lung cancer based on network pharmacology

摘要 目的运用网络药理学方法预测十全育真汤治疗非小细胞肺癌(NSCLC)的作用机制。方法检索中药系统药理学数据库与分析平台(TCMSP)、PubMed等相关数据库,筛选出十全育真汤的活性成分及对应靶点;通过GeneCards等疾病数据库筛选出NSCLC相关靶点;将药物靶点与疾病靶点相映射得到十全育真汤治疗NSCLC的靶点;采用Cytoscape 3.7.2软件筛选出关键活性成分,并构建"药物-靶点-疾病"网络;采用DAVID 6.8数据库分析基因本体(GO)及京都基因与基因组百科全书(KEGG)信号通路;采用STRING V11.0数据库构建蛋白质-蛋白质相互作用(PPI)网络。结果筛选出十全育真汤治疗NSCLC的关键活性成分26个,包括槲皮素、木犀草素和山柰酚等,关键活性成分对应的靶点182个;GO功能及KEGG富集分析显示,信号通路主要涉及磷脂酰肌醇3激酶/蛋白激酶B(PI3K-Akt)信号通路、肿瘤坏死因子(TNF)信号通路、缺氧诱导因子-1(HIF-1)信号通路和血管内皮生长因子(VEGF)信号通路等15条通路;PPI网络分析显示,十全育真汤治疗NSCLC的核心靶点主要集中在细胞增殖、凋亡、细胞周期、炎症及血管生成等方面。结论十全育真汤治疗NSCLC的作用机制可能与提高免疫、抑制血管生成及促进细胞凋亡等有关。 Objective To use network pharmacology for predicting the action mechanism of Shiquan Yuzhen Tang( 十全育真汤) in the treatment of non-small cell lung cancer( NSCLC). Methods Traditional Chinese Medicine Systems Pharmacology( TCMSP),Pub Med and other related databases were searched to screen out the active components and corresponding targets of Shiquan Yuzhen Tang;NSCLC related targets were screened out by GeneCards and other disease databases;The targets of Shiquan Yuzhen Tang in the treatment of NSCLC were obtained by mapping the drug targets with the disease targets;The key active components were screened out by Cytoscape 3.7.2 software,and the "drug-target-disease"network was constructed;DAVID 6.8 database was used to analyze gene ontology( GO) and Kyoto Encyclopedia of Genes and Genomes( KEGG) signal pathway;The protein-protein interaction( PPI) network was constructed using STRING V11.0 database. Results Twenty-six key active components of Shiquan Yuzhen Tang in the treatment of NSCLC were screened out,including quercetin,luteolin and kaempferol,and 182 targets corresponding to key active components;GO function and KEGG enrichment analysis showed that the signal pathways mainly involved 15 pathways,namely phosphatidylinositol 3 kinase/protein kinase B( PI3 KAkt) signal pathway,tumor necrosis factor( TNF) signal pathway,hypoxia inducible factor-1( HIF-1) signal pathway and vascular endothelial growth factor( VEGF) signal pathway,etc.;PPI network analysis showed that the core targets of Shiquan Yuzhen Tang in the treatment of NSCLC mainly focused on cell proliferation,apoptosis,cell cycle,inflammation and angiogenesis,etc. Conclusion The action mechanism of Shiquan Yuzhen Tang in the treatment of NSCLC may be related to improving immunity,inhibiting angiogenesis and promoting apoptosis.
