摘要
为了提高口蹄疫(FMD)灭活疫苗的免疫效果,用由Zn2+和2-甲基咪唑配位而成的ZIF-8(沸石咪唑酯骨架)包封灭活的FMDV,探究其对免疫效果的影响。最终成功制备出粒径约为154 nm的ZIF-8-FMDV复合物。组织毒性试验证明,该复合物具有较好的生物相容性。与未包封的灭活的FMDV相比,该复合物诱导豚鼠产生的特异性抗体水平显著升高,并提供较高的攻毒保护率。结果表明,用ZIF-8包封FMDV可增强病毒抗原的免疫效果,为通过化学、材料学等交叉学科方法优化抗原及提升疫苗的免疫原性提供了新的思路。
To enhance the immune effect of the foot-and-mouth disease(FMD) inactivated vaccine,inactivated foot-and-mouth disease virus(FMDV) was encapsulated with ZIF-8 coordinated by Zn2+ and 2-methylimidazole and explored its impact on the immune effect.Here,the ZIF-8-FMDV with a size of about 154 nm was successfully prepared.Tissue toxicity test showed that the compound has excellent biocompatibility.Compared with the inactivated FMDV,the complex induced significantly higher levels of the specific antibodies and improved protection against challenge in guinea pigs.These data indicated that encapsulation of inactivated FMDV by ZIF-8 can enhance its immune effect,which also provides innovative for optimizing antigens and improving the immunogenicity of vaccines through interdisciplinary methods such as chemistry and materials science.
