摘要
作为有效抵御DNA病毒感染的方式,环腺苷酸鸟苷酸合成酶(cyclic GMP-AMP synthase,cGAS)及其下游干扰素刺激基因(stimulator of interferon genes,STING)在宿主细胞中通过识别DNA分子,可级联放大反应调节宿主干扰素(interferons,IFNs)及促炎细胞因子的表达,从而发挥抗DNA病毒感染作用。针对cGAS-STING抗病毒感染的信号通路,许多DNA病毒也已经进化出多种拮抗策略,通过作用于该通路中不同的蛋白分子逃逸宿主的免疫应答。本文就cGAS-STING通路识别DNA病毒并诱导宿主天然免疫反应以及DNA病毒拮抗cGAS的识别策略的研究进展作一综述,以期为深入理解DNA病毒与宿主之间的相互作用奠定基础,也为基于cGAS-STING通路为靶点的疫苗研发及药物设计提供重要的理论支撑。
As a way to effectively defend against DNA virus infections,cyclic GMP-AMP synthase(cGAS) and its downstream stimulator of interferon genes(STING) in host cells identified the DNA molecules,and adjusted the host interferons(IFNs) and proinflammatory cytokine gene expression through the cascade amplification reaction,and thus played an anti-DNA-viral role.Many DNA viruses had also evolved a variety of antagonistic strategies targeting the signaling pathway of cGAS-STING antiviral infection,which acted on different protein molecules in the pathway to escape the host immune response.This paper reviewed the research progress of the cGAS-STING pathway recognizing DNA viruses and inducing host innate immune response,as well as the recognition strategies of DNA viruses antagonizing cGAS.This will help to lay a foundation for further understanding the interaction between DNA viruses and hosts,and also provided important theoretical support for the research and development of vaccines and drug design based on cGAS-STING pathway.
