摘要
为了分析18份辣木育种亲本间的亲缘关系,以期为辣木杂交育种、分子标记辅助育种提供理论支持,本研究从40条SCoT引物中筛选条带清晰、多态性好、杂带少的引物,应用SCoT分子标记对18份辣木育种亲本进行PCR扩增,统计分析电泳图谱,利用NTSYS 2.1软件计算遗传相似系数,通过SHAN程序进行UPGMA聚类分析,并绘制出树状聚类图。研究结果表明,从40条SCoT引物中筛选出了14条多态性丰富的引物,并扩增出清晰条带114个,其中具有多态性的条带有78个,平均每条引物获得多态性条带5.57个,多态性比例达68.42%;基因多样性指数(H)为0.266 1,有效等位基因数(Ne)为1.470 9,Shannon信息指数(I)为0.388 5;UPGMA聚类分析结果表明,18份辣木育种亲本的遗传相似系数为0.61~0.96;在遗传相似性系数0.61水平上,18份辣木可分为两大类群,来源于非洲卢旺达的狭瓣辣木自成一类;系数为0.732处,其余种质资源又聚为两大类群。SCoT标记能较好地反映辣木育种亲本间的遗传关系,辣木育种亲本种质资源间具有较丰富的遗传多样性。
In order to provide theoretical support for hybrid breeding and molecular marker assisted breeding, the present study analyzed the genetic relationship among 18 Moringa spp. breeding parents. SCoT primers with clear amplified bands, few heterobands and rich polymorphism were selected from 40 primers. The DNA of 18 breeding parents of Moringa was amplified by PCR. The electrophoretogram was analyzed, and the SM similarity coefficient was calculated by NTSYS 2.1 software. Then the UPGMA clustering analysis was carried out with SHAN program to draw the dendrogram. The results showed that 114 clear bands were amplified by 14 primers with high polymorphism, among which 78 were polymorphic bands, with an average of 5.57 polymorphic bands per primer,and the average percentage of polymorphic bands was 68.42%. The genetic diversity index(H), effective allele number(Ne) and Shannon’s information index(I) were 0.266 1, 1.470 9 and 0.388 5, respectively. UPGMA cluster analysis showed that the genetic similarity coefficient of 18 breeding parents of Moringa spp. was between 0.61~0.96. At the level of genetic similarity coefficient of 0.61, the samples could be divided into two groups, and the samples of Moringa stenopetala from Rwanda, Africa were grouped into one group;at the coefficient of 0.732, the other germplasm resources were grouped into two groups. The result revealed that SCoT molecular markers are suitable for the analysis of genetic relationship of the Moringa spp. breedings. The genetic diversity of the Moringa spp. breeding germplasm resources is quite rich.
