摘要
为了研究蜜蜂巢脾多糖的免疫调节活性,试验设置空白对照(K)组、模型对照(M)组、阳性对照(Y)组及蜜蜂巢脾多糖低剂量(F1)组、蜜蜂巢脾多糖中剂量(F2)组、蜜蜂巢脾多糖高剂量(F3)组,对除K组外的各组小鼠腹腔注射环磷酰胺构建免疫抑制模型,采用qRT-PCR技术检测各组小鼠脾脏细胞因子IL-4、IFN-γ及Th1/Th2转录因子T-bet和GATA-3 mRNA的表达。结果表明:相比K组,M组Th2转录因子GATA-3 mRNA及细胞因子IL-4 mRNA的表达显著提高(P<0.05),而Th1转录因子T-bet mRNA及细胞因子IFN-γmRNA的表达显著降低(P<0.05)。与M组相比,Y组及F2组Th1转录因子T-bet mRNA及细胞因子IFN-γmRNA的表达均显著提高(P<0.05),而Y组及蜜蜂巢脾多糖各组中Th2转录因子GATA-3 mRNA及细胞因子IL-4 mRNA的表达虽有所降低,但与M组差异不显著(P>0.05)。说明蜜蜂巢脾多糖可通过上调Th1转录因子T-bet mRNA及细胞因子IFN-γmRNA的表达促进免疫抑制小鼠机体Th1/Th2恢复平衡。
In order to research the immunomodulatory activity of honeybee comb polysaccharide, the experiment set up blank control(K)group, model control(M)group, positive control group(Y),honeybee comb polysaccharide low-dose(F1)group, honeybee comb polysaccharide medium-dose(F2)group, honeybee comb polysaccharide high-dose(F3)group, and immunosuppressed mouse model was prepared by intraperitoneal injection of cyclophosphamide except K group. The qRT-PCR technique was used to detect the mRNA expression of spleen cytokines IL-4,IFN-γ,Th1/Th2 transcription factors T-bet and GATA-3 in each group of mice. The results showed that compared with K group, the mRNA expression of Th2 transcription factor GATA-3 and cytokine IL-4 was significantly increased(P<0.05) in M group, while the mRNA expression of Th1 transcription factor T-bet and cytokine IFN-γ was significantly decreased(P<0.05). Compared with M group, the mRNA expression of Th1 transcription factor T-bet and cytokine IFN-γ was significantly increased(P<0.05) in Y group and F2 group, while the mRNA expression of Th2 transcription factor GATA-3 and cytokine IL-4 was decreased in Y group and honeybee comb polysaccharide groups, which had no significant difference compared with M group(P>0.05). The results suggested that honeybee comb polysaccharide could up-regulate the mRNA expression of Th1 transcript factor T-bet and cytokine IFN-γ,for the promotion of the balance of Th1/Th2 in immunosuppressed mice.
