川西高原垂穗披碱草根际土壤放线菌分离与鉴定 被引量:1

Isolation and Identification of Actinomycetes from Rhizosphere Soil of Elymus nutans in Western Sichuan Plateau

摘要 为挖掘川西高原植物微生物资源,完成微生物资源的分离及鉴定,本研究通过采集川西高原垂穗披碱草根际土壤样本,实验将对分离后的放线菌进行形态特征观察、生理生化鉴定、16S rRNA基因扩增比对以及系统发育树的构建以确定其种属情况,并通过牛津杯法测定菌株发酵液的抑菌活性。结果表明,从川西高原选择的6个土壤代表区共分离纯化得到5株具有抑菌活性的菌株;在抑菌活性的实验中,5株菌株表现出对病原菌有一种或者多种抑制的特性。经过对菌株进行生理生化鉴定、16S rRNA基因扩增比对以及系统发育树的构建,结果显示5株菌株均属于链霉菌属(Streptomyces)。 In order to excavate the plant microbial resources in western Sichuan plateau and to complete the isolation and identification of the microbial resources,the present study collected samples from the rhizosphere soil of the Elymus nutans in the western Sichuan Plateau.The species of the isolated actinomycetes are determined by morphological observation,physiological and biochemical identification,16S rRNA gene amplification,and phylogenetic tree construction,the antibacterial activity of the fermentation broth of the strain against four pathogenic bacteria is determined by Oxford Cup method.The results show that 5 strains with bacteriostatic activity are isolated and purified from 6 soil representative areas in the Western Sichuan Plateau.In the experiment of bacteriostatic activity,5 strains shows one or more kinds of inhibition to pathogenic bacteria.After physiological and biochemical identification,16S rRNA gene amplification and phylogenetic tree construction,the results shows that all 5 strains belonged to Streptomyces.
