HPLC法测定蒲黄镇痛活性提取物的含量

Determination of Analgesic Components in Pollen Typhae Extract by HPLC

目的:建立HPLC法测定蒲黄镇痛提取物中香蒲新苷的含量,为蒲黄镇痛成分的质量控制与评价奠定基础。方法:色谱柱采用Amethyst C18-H(4.6 mm×250 mm,5μm),流动相为乙腈-0.05%磷酸(15∶85),UVD检测波长为254 nm,流速为1.0 mL/min,柱温为25℃,进样量为20μL。结果:蒲黄镇痛成分中香蒲新苷在20~100μg/mL的浓度范围内线性关系良好(R^(2)=0.9972);加样回收率为103.42%,RSD值(n=6)为1.16%。结论:建立HPLC法测定蒲黄镇痛提取物中香蒲新苷的含量,有效可靠,蒲黄镇痛提取物中香蒲新苷的含量为0.64%。研究为蒲黄镇痛成分的质量控制与评价奠定了基础。

Objective:To establish a HPLC method for the determination of typhaneoside in the analgesic components of Pollen Typhae extract.Methods:The chromatographic conditions are:Amethyst C18-H(4.6 mm×250 mm,5μm)on a chromatographic column,mobile phase is acetonitrile-0.05%phosphoric acid(15∶85),UVD detection wavelength 254 nm,flow rate 1.0 mL/min,column temperature 25℃,amount of injection 20μL.Results:There is a good linear relationship between in the analgesic components of typhaneoside(R^(2)=0.9972)within the concentration range of 20~100μg/mL.The average recovery rate is 103.42%and the RSD value(n=6)is 1.16%.Conclusion:HPLC method is established to determine the content of typhaneoside in the extracts of analgesic extracts.It is effective,reliable and accurate.The content of typhaneoside in the analgesic extracts is 0.64%.
