基于TGF-β1、EGF、TNF-α水平研究吉西他滨对膀胱炎模型大鼠灌注的作用

摘要 目的研究膀胱灌注吉西他滨对膀胱炎模型大鼠转化生长因子-β1(TGF-β1)、表皮细胞生长因子(EGF)、肿瘤坏死因子-α(TNF-α)水平的影响。方法将30只雌性大鼠随机分为吉西他滨组、模型组和对照组,吉西他滨组和模型组灌注DH5α大肠杆菌溶液,建立腺性膀胱炎大鼠模型,吉西他滨组灌注吉西他滨1.0 g+50 ml 0.9%的氯化钠液体,对照组灌注等量0.9%氯化钠注射液。随后对三组大鼠进行尿动力学检测,并用荧光定量PCR(RT-PCR)和免疫蛋白印迹法对TGF-β1、EGF、TNF-αmRNA及其蛋白进行检测。结果吉西他滨组、模型组的最大膀胱测压容积、排尿间隔、膀胱顺应性低于对照组(t分别=8.34、9.91、7.90;1.76、3.93、3.49,P均<0.05),吉西他滨组的最大膀胱测压容积、排尿间隔、膀胱顺应性高于模型组(t分别=6.20、5.32、6.32,P均<0.05);吉西他滨组、模型组TGF-β1、EGF、TNF-α基因mRNA的相对表达量高于对照组(t分别=2.93、3.34、3.52;6.72、6.71、5.81,P均<0.05),与模型组比较,吉西他滨组的TGF-β1、EGF基因mRNA的相对表达量较高,TNF-α基因mRNA的相对表达量较低(t分别=3.64、3.19、2.08,P均<0.05);吉西他滨组、模型组TGF-β1、EGF、TNF-α蛋白的相对表达量高于对照组(t分别=8.76、5.81、4.65;8.76、11.61、1.70,P均<0.05),与模型组比较,吉西他滨组的TGF-β1、EGF蛋白的相对表达量较高,TNF-α蛋白的相对表达量较低(t分别=2.80、5.80、2.79,P均<0.05)。结论灌注吉西他滨能够有效治疗腺性膀胱炎,其作用机制可能与吉西他滨对TGF-β1、EGF和TNF-α的调控有关。 Objective To study the effect of gemcitabine infusion on transforming growth factor in cystitis model ratsβ1(TGF-β1),epidermal growth factor(EGF),tumor necrosis factorα(TNF-α)levels in cystitis model rats.Method The 30 female rats were randomly divided into the gisitamine group,model group and control group.The gissitamine group and the model group perfused DH5 alpha E.coli solution,established adenoid cystitis rat model,and the gisitamine group perfusion of gisitabin 1.0 g plus 50 ml 0.9%sodium chloride liquid,the control group perfusion and other physiological saline.Then urodynamics of three groups were detectea,and the relative expression of TGF-β1,EGF,TNF-αmRNA and their proteins were detected by fluorescent quantitative PCR(RT-PCR)and Western blotting.Results The maximum bladder pressure measurement volume,voiding interval,and bladder compliance of the gemcitabine group and model group were lower than those of the control group(t=8.34,9.91,7.90,1.76,3.93,3.49,P<0.05),the maximun bladder pressure measurement volume,voiding interval,and bladder compliance of the gemcitabine group were higher than those of the model group(t=6.20,5.32,6.32,P<0.05),the relative expression of TGF-β1,EGF,and TNF-αgene mRNA in the gemcitabine group and model group were higher than those of the control group(t=2.93,3.34,3.52,6.72,6.71,5.81,P<0.05),compared with the model group,the relative expression of TGF-β1 and EGF gene mRNA in the gisitabin group was higher,and the relative expression of TNF-αgene mRNA was lower(t=3.64,3.19,2.08,P<0.05).The relative expression of TGF-β1,EGF and TNF-αprotein in gemcitabine group and model group were higher than those of control group(t=8.76,5.81,4.65,8.76,11.61,1.70,P<0.05),compared with the model group,the relative expression of TGF-β1 and EGF protein in the gisitabin group were higher,and the relative expression of TNFαprotein was lower(t=2.80,5.80,2.79,P<0.05).Conclusion Gemcitabine infusion can effectively treat glandular cystitis,and its mechanism may be related to the effect of gemcitabine on TGF-β1,EGF and TNF-αrelated to the regulation.
