达格列净的合成方法

摘要 对达格列净的合成方法进行改进。以5-溴-2-氯-4′-乙氧基二苯甲酮为原料,在三乙基硅烷/三氟化硼乙醚条件下发生还原反应得到5-溴-2-氯-4′-乙氧基二苯甲烷,5-溴-2-氯-4′-乙氧基二苯甲烷与苄基保护的葡萄糖内酯、正丁基锂利用微通道反应技术发生交叉偶联反应生成糖苷键,再用三乙基硅烷和三氟化硼乙醚还原脱除羟基,最后在Pd/C和甲酸铵的催化下脱除苄基得到达格列净纯品,所得产物的总收率67%,纯度为99.12%。该路线安全性较低的正丁基锂参与的交叉偶联反应采用微通道反应技术,可提高安全性,葡萄糖利用苄基保护,避免羟基的多次衍生化和杂质的产生,改进后的方法反应步骤少、操作简便、安全性高、立体选择性高,可用于达格列净的放大合成。
