葡萄球菌脂磷壁酸对糠秕马拉色菌诱导的HaCaT细胞IL-6及IL-1β的影响 被引量:1

摘要 目的明确脂磷壁酸(LTA)对糠秕马拉色菌诱导的HaCaT细胞白细胞介素(IL)-6及IL-1β影响。方法将HaCaT细胞分为LTA与糠秕马拉色菌共处理组、糠秕马拉色菌处理组和对照组。在共培养后的不同时间点,应用实时荧光定量聚合酶链反应(Rt-PCR)和酶联免疫吸附测定(ELISA)法检测不同组别HaCaT细胞IL-6及IL-1β的表达。结果糠秕马拉色菌与HaCaT细胞共培养后1 h,糠秕马拉色菌可诱导HaCaT细胞IL-6及IL-1βmRNA表达升高。LTA和糠秕马拉色菌共同处理组HaCaT细胞IL-6及IL-1βmRNA的表达低于糠秕马拉色菌处理组。与正常培养的HaCaT细胞上清相比,糠秕马拉色菌促进HaCaT细胞分泌IL-6。LTA和糠秕马拉色菌共同处理组上清IL-6分泌低于糠秕马拉色菌处理组。结论葡萄球菌LTA可以抑制糠秕马拉色菌诱导的炎性因子IL-6及IL-1βmRNA表达,抑制糠秕马拉色菌诱导IL-6分泌。 Objective In order to investigate the effects of staphylococcal lipoteichoic acid(LTA)on the expression of interleukin(IL)-6 and IL-1βinduced by Malassezia furfur(M.furfur)in HaCaT cells.Methods HaCaT cells were divided into three groups:LTA and M.furfur co-treatment group,M.furfur treatment group and control group.At various time-points,real-time polymerase chain reaction(RT-PCR)and enzyme-linked immunosorbent assay(ELISA)were used to evaluate the effects of Malassezia strains on cytokine expression.Results M.furfur could induce HaCaT cells IL-6 and IL-1βmRNA to increase the expression after co-culturation of M.furfur and HaCaT cells for 1 h.The expression of HaCaT cells IL-6 and IL-1βmRNA was higher in LTA and M.furfur co-treatment group than that of M.furfur treatment group.It was found in detection of cytokines in cell cultured supernatant using ELISA method that M.furfur could induce HaCaT cells to secrete IL-6,compared to normal cultured group.Conclusion Staphylococcal LTA can inhibit stimulation of M.furfur,the expression of IL-6 and IL-1βmRNA in HaCaT cells is decreased and secretion of IL-6 is reduced.
